Study On The SNPs Of Ex-FABP Gene And Its Association With The Fatty Traits In Chickens

In this study, DNA sample were extracted from 128 chickens including four Chinese native chicken breeds : Qingyuan Ma chicken, Huiyang Huxu chicken, Fengkai Xinghua chicken, Guangxi Xiayan chicken and three specific strains : Lingnan Huang fast-growing- II chicken, Lingnan Huang with spotting feather and draw gene and Arbor Acres (imported line) . Ex-FABP gene was studied by PCR -SSCP with seven pair primers. Each homozygote found in SSCP of Ex-FABP gene was sequenced. Meanwhile the effect of different genotypes on the rate of abdominal fat and IMF (Intra-muscular Fat, IMF) contents were analyzed. The aim of this study was to determine the major gene affecting the rate of abdominal fat and IMF content or the genetic markers tightly linked with it.The Ex-FABP gene in the seven chicken breeds (stains) didn't test out the SNP sites by PCR-SSCP in the PCR products of primer P4 and P6. The SSCP results of the PCR products of primer P1, P2, P3 and P7 showed that there were three genotypes.The SSCP results of primer P2 showed that the AA genotype was not found in Arbor Acres and the BB genotype was the most common genotype.The SSCP results of primer P3 showed that there was only AB genotype found in Lingnan Huang fast-growing-II chicken, Huiyang Huxu chicken and Fengkai Xinghua chicken, but all the three genotypes-AA, BB and AB were found in Arbor Acres, Lingnan Huang with spotting feather and draw gene and Qingyuan Ma chicken. At the same time, the frquences of three genotypes in the Lingnan Huang with spotting feather and draw gene and Qingyuan Ma chicken were the same.The SSCP results of primer P7 showed that there were three genotypes-AA, AB and AB in the all chickens straits were tested in this study, and the AB genotype were common in Chinese native breeds and Lingnan Huang fast-growing- II chicken.The result of primer P5 showed that there were four bond-types-â… , â…¡ ,â…¢ and â…£ in...
