Prunella vulgaris L., a traditional Chinese herb which mainly grows in wild has many medical functions such as detumescence, acesodyne, et al. As one of the major product provinces Sichuan has plenty of Prunella vulgaris L.. By plant biotechnology and chemical methods, this paper studies on biodiversity and chemical elements of Prunella vulgaris L., which will provide basis for medical quality standardization of Prunella vulgaris L..In the experiments, two kinds of molecular markers technology on Prunella vulgaris L. were built up, i.e. rapid amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). The better technology was applied to study the genetic diversity analysis of Prunella vulgaris L.. The percentage of ursolic and oleanolic Acids can be determined at the same time with the reversed-phase high performance liquid chromatography (HPLC). The differences in acid percentage were found resulting from the different collect time and process parts. Furthermore, tissue culture of Prunella vulgaris L. was carried out from their explants. The growth, root generation and proliferation of the tissue culture were observed and recorded. Combined with the effective percentage test results, the mediums which were fit for the tissue culture of Prunella vulgaris L. were proposed for the further experimental.It was found that, the variation of Prunella vulgaris L. tested by RAPD technology is too small to continue the subsequent cluster analysis. However, AFLP technology has high test efficiency, fine precision and well repeatability, which satisfies the experimental requirements. It could be observed to have 682 DNA bands...
