Bulblets of lily-Aziatics of two species Nove Cento,Brunello and leaves in bulblet ofNove Cento were selected for in vitro culture.Five stages of primary research wereprocessed,that were,establishment of clone system,multiplication of embryonal callus,multiplication of sprout, rooting and bulblet formation and transplantation.The resultare shown as follows:1. Stage of establishment of clone systemThe bulblets of two species, Nove Cento and Brunello ,were selected as explants forin vitro culture,and compared their differentiation rate and contamination rate.Theresults of experiments indicated that the Nove Cento differentiation rate was raised asthe concentration of hormone was raised.Among the inducing mediums in ourexperiment,MS medium supplemented with 3.0mg/L 6-BA and 0.1mg/L NAA was thebest for Nove Cento,its inducing rate was 82.4 percent;While the Brunellodifferentiation rate was droped as the concentration of hormone was raised, MSmedium supplemented with 1.0mg/L 6-BA and 0.2mg/L NAA was the best forBrunello ,its inducing rate was 84.6 percent.The leaves in bulblet of Nove Cento were selected as explants for in vitro culture.The results of experiments indicated that the time of differentiation was short,differentiation rate and coefficient of differentiation were high. Among the inducingmediums in our experiment, MS medium supplemented with 2.0mg/L 6-BA and 0.2mg/LNAA was the best.The basal part of bacteria and grem-free leaves of two species, Nove Cento andBrunelio, were selected as explants for in vitro culture. The results of experimentsindicated that MS medium supplemented with 2.0mg/L 6-BA and 0.1mg/L NAA was thebest for Nove Cento,while MS medium supplemented with 2.0mg/L 6-BA and 0.2mg/LNAA was the best for Brunello. 2. Stage of multiplication and differentiation of embryonal callusDuring the stage of embryonal callus of multiplication and differentiation,it wasfound that,2.0~2.5mg/L 6-BA was the better range for multiplication anddifferentiation. Among the inducing mediums in our experiment,MS mediumsupplemented with 2.5mg/L 6-BA and 0.2mg/L NAA was the best for Nove Cento,whileMS medium supplemented with 2.0mg/L 6-BA and 0.1mg/L NAA was the best forBrunello..3.Stage of sprout multiplicationDuring the stage of sprout multiplication, it was found that, 6-BA and NAA withlow concentration(0.1~0.2mg/L) can promote sprout multiplication. The multiplicationquotiety of Nove Cento was higher, MS medium supplemented with3.0mg/L 6-BA and0.2mg/L NAA was the best; The multiplication quotiety of Brunello was lower, MSmedium supplemented with2.0mg/L 6-BA and 0.2mg/L NAA was the best.4.Stage of rooting and bulblet formationDuring the stage of rooting and bulblet formation,the result showed that theconcentration of sugar and macroelement, affected rooting and forming ofbulblets.Sucrose can help to form bulblet , the best concentration was 60g/L.It wasfavorable for rooting and bulblet formation by reducing the concentration ofmacroelement.3/4MS proved the concentration for Nove Cento,while 1/2MS proved theconcentration for Brunello.5. Stage of transplantationThere were four stages in transplantation,that sand bed prepairing,domestication,pseudoplantation and transplant soil and basin.The survival rate oftransplantation can reach 100precent in the highest level.
