Populus is one of an important industrial tree.There are many researches about tissue culture and rapid propagation of Populus, mostly focusing on native variety. Using stem sections of triploid European aspen as testing material. We study on the influencing factors, sampling time, sterilization, rapid propagation, rooting and transplant included. A preliminary rapid propagation system of triploid European aspen is established, and it is of great significance for the rapid propagation and extension of fine tree.In mid April we use biennial stem sections as material, and culture them in wet sand. When buds grow big enough, we cut off growing points,petioles and laminae as explants.In sterilization process step, explant is put in soapy water for 10 minutes, next scoursed them for 3 hours in running water, then dip them in 75% alcohol and 0.1% mercuric chloride separately for 30 seconds and 6 minutes, at last use sterilization water washing them 3 to 5 times. The sterilizton effecfs is good. We study on the type of explants,basic medium and the kinds and concentration of plant growth regulator, the results show that the highest inducing rate is petiole explant cultivated in MS basic medium added with 0.5 mg/l 6-BA and 0.05 mg/l NAA, up to 580.36%. We study on rooting inducement of adventitious buds cultivated on 1/2MS basic medium added with different concentration of NAA and IBA. The results show that when adventitious buds are cultivated in 1/2MS basic medium added with only 0.25 mg/l IBA, rooting rate is the best, up to 97.50%. The plantlets are transplanted into medium which is formed by soddy soil and farm soil, the constitutional ratio is 1:1. The plantlets are shaded for 3 days in greenhouse in which temperature is 24 to 28 degrees Celsius before cultivated under natural light for 20 days. Then they are transplanted in natural conditions, and the survival rate is to 86.23%.
