Study On The Optimum Conditions In Liquid Culture And Man Mutation Breeding Of Cordyceps Militaris (L) Link

Cordyceps militaris(L.)Link is one of the most important fungus in condyceps genus ,as the Cordyceps sinensis is .It can live in shallow hill and some kinds of insects ,while Cordyceps sinensis live in deep mountain and a few kinds of insects .We can get hypha by liquid culturing and can get kernel by solid culturing. It can replace the famous Cordyceps sinensis in diet and traditional medicine in Chinese.Some ingredients in it is as much as the wild Cordyceps,uch such as CMP and C7H12O6.Some others ingredients in it is as 35 times as the wild's,such as C10H13O3N5.Its effective activity ingredients CMPS is regarded as a kind of substance that can prevent you from lots of disease.It can make the immune cell in the body active.But wild Cordyceps is so scarce that it can not be used as wide as we expect.Man-made cultivating especially liquid cultivating can make Cordyceps militaris(L.)Link be used as wide as we expect.Nowaday man-made cultivating is used widely more and more as the science developing because the new use of it.Some ingredients in Cordyceps militaris(L.)Link such as CMPS and C7H12O6 are needed increasing .The study is about separating tissue ,cultivating new fungus through liquid fermentation and breaking the hypha and using it for mutation breeding .Thus we can find out new kinds cordyceps militaris from the mutation breeding hypha through checking on some quality of them .It bases applyment on theory .This study proceed from Cordyceps militaris(L.)Link . lead the pieces fungus to mutate Using ultraviolet ray or microwave and two of them.Find out that the longer time we shine it ,the little seed can survive .and use the CMPS in them and the weight of different hypha as index .Meantime do experiment to verify the new hypha's inheritance steady through 4 generation cultivating .It show that the inheritance is stable .Through single factor and orthogonal design method , the shaking culture experiment results show that the optimum conditions in liquid culture for 5d are:peptone1.5%,cane sugar2%, KH2PO40.1%,MgSO40.1%,20℃,PH7, 170r/min.Under this condition ,the CMPS in new Cordyceps militaris(L.)Link is 30.4243mg and 28.9692mg in original Cordyceps militaris(L.)Link .Show that it is 5% higher than before .At the same time, solid cultivating experiment results show that the CMPS in new Cordyceps militaris(L.)Link is 3.2713mg/g and 2.7955 mg/g in original Cordyceps militaris(L.)Link . Show that it is 17% higher than before under the same condition .It is worth to apply.The innovative aspect of the study is to try to use the pieces fungus for mutation breeding for the first time, including using ultraviolet ray or microwave and two of them.
