Guzmania lingulata,a kind of indoor foliage and flowers viewing flower , has high ornamental value and was appreciated by people because of six-month florescence. Stem, tender leaf sheath and old leaf sheath were used as a source of initial explants in this study. We studied on initiation culture, the bud induction, rooting. After proliferation, rejuvenation, domestication and transplant, it had achieved rapid breeding of Guzmania lingulata purpose. The results of the study will provide a solid theoretical foundation on clones of breeding, genetic preservation, factory production areas. The mains results are as follows:1. The best method for sterilize of explants: it was dip in 70% ethyl alcohol for 30s, then agitating in a solution of 0.1%HgCl2 for 8 min for stem tender leaf sheath and old leaf sheath, finally rinsed five times with sterile distilled water.2. The effect of different selected material on the callus induction is different. The old leaf sheath for the callus rate is the highest, followed is the stem, but the tender leaf sheath is the lowest.3. The medium: the best basic medium for callus induction was MS. and the suitable growth hormone combination and density was 6-BA2.0mg/L+NAA0.2mg/L. With hormone combination and density 6-BA2.0mg/L+NAA0.25mg/L,the bud could grow well and accelerate the multiplication. And with 8mg/L coconut milk, the bud can get stronger.4. The glucose was better than the sucrose to the callus induction .The rate of callus induction with the glucose was obviously higher than the sucrose.5. Illumination is better than darkness with the callus induction and the bud growth, and with strength of illumination for 1500lux-2000lux natural diffused light was better. But the darkness was bad for the callus induction and the bud growth.6. The rate of callus induction with the 3 different varieties for Guzmania lingulata was different .It possible concerns with the gene of the variety.7.In subculture, choosing shoots to inoculate on the medium, which make the effect of multiplication, was evident.8 .In subculture, the more of the subculture time, the higher of coefficient of multiplication. The coefficient increased rapidly in five generation, and then the coefficient not increased more like before or decreased after five generation. 9 .On medium supplied CM 8mg/L, CM can promote growth of shoots, and make effective rejuvenation cultivation.10 .The best medium for rooting was 1/2MS+NAA0.5mg/L.The quality of the root system was better than others, and the rooting rate was 100%. The survive ratio could reach 96% when they were transplant to the medium composed with one part perlite, two part coconut tree rice polishings and one part muck soil .11 After the adventitious bud were rejuvenated, may overcome social buds breeding coefficient to be reduced and its quality becoming unstable after continues 5 generations Thus not only may provide the neat consistent vigorous and healthy adult plant, moreover may carry on the multi- generations breeding, provides the circulation system for the factorization quick numerous production which may operate.
