On the basis of a large number of Chinese and foreign references, the fingerprint analysis of Coptis deltoidea C.Y.Cheng et Hsiao has been researched and established by high performance liquid chromatographic technology. That has been because of the actual demands, not only for insuring the reputation of Coptis deltoidea C.Y.Cheng et Hsiao produced in Emei ,but for the rescue of Yanlian produced in Emei which is in severe danger and at the same time in order to plant widely as well. At this time, it has provided the gist to constitute the quality standard of Coptis deltoidea C.Y.Cheng et Hsiao.This paper consists of two chapters:In chapter one, review the contents as followed:The summary of fingerprint analysis of Chinese traditional medicine; The methods of establishment of fingerprint and its applications; The sixty-four-dollar questions in the process of its research; The issue and expectation of fingerprint analysis of Chinese traditional medicine.In chapter two, set up the fingerprint analysis of Coptis deltoidea C.Y.Cheng et Hsiao by high performance liquid chromatographic technology.On the basis of a great deal of chromatographic data, the fingerprint analysis of Coptis deltoidea C.Y.Cheng et Hsiao was established and decoded, by using high performance liquid chromatographic technology and uniting a series of high performance technology of extraction and separation. All the contents included the preparation for the samples and the standard, the determination to the conditions of elution and the choice of detection wavelength, column temperatures, injection quantity and etc. The examination of methodologies was also research , which included the experiments of stability, precision and fidelity.At last, the optimal conditions were determined by a large number of experiments. The best solvent to extract alkaloids is HCl-methanol(1:100). At 40℃and the injection of 10μl, using the solution of 0.05moL/L potassium dihydrogen phosphate and acetonitrile at different proportions as the elution, the optimal detection wavelength is 345nm.The results of the examination of methodologies showed that the RSD of relative time of each peak was less than 0.5%, and the RSD of the relative area of the peak was less than 3.0%.
