This study was focus on the selection of culture matrix for calla's culture.Based on the experiment in vitro, the rule of physical alteration which belongs to the calla in the course of its decline has been studied.The best growth performance and flower qualities were observed for the growth of the two varieties in substance peat, than in matrix T4(the mixture of peat, mushroom debris and sawdust in the volume tatio of 1:1:1) or T6(the mixture of peat, mushroom debris and silver sand in the volume tatio of 1:1:1).In the course of calla's decline, the peroxydase(POD),catalase(CAT),the content of soluble protein, percentage of moisture of calla's cut flower rise formerly and decrease latter. the content of propylene glycol(MDA), cell membrane permeability and the content of superoxide radical(O2-) rise consistently. The pattern of the change of superoxide dismutase(SOD)'s activeness is similar with the shape of letter "M". Among the physical alteration mentioned above, some are responsible for the decline of cut flower, others result from the decline of cut flower which are the gist of developing antistaling agent.According to the study, the specilized antistaling agent for calla's cut flower is the aqueous solution which contains 0.5% cane sugar+ 0.5% polyoxyethylene lauryl ether + 100mg/l lauryl xylidine sal ammoniac+ 10mg/l 6-benzyl aminopurine (6-BA) +100mg/l Co(NO3)2 which can prolong the time 216h, the percentage of delay is 112.5%. In general, The activeness of POD,CAT and SOD can be grown, the decline of the content of soluble protein and of the percentage of moisture of calla's cut flower, the rise of the content of superoxide radical, of MDA and of the cell membrane permeability can changed more slowly with the effection of antistaling agent developed by this study.
