Colored calla lily(Zantedeschia hybrida)is a bulbous flower with good ornamental quality.It is deeply loved by consumers in china and abroad and has a reputation of “colored calla lily”.The colored calla lily flowers which are current on the market are mostly cultivated in New Zealand,the Netherlands and the United States.Comparing with these countries,the breeding of new varieties of calla lily in China start late and there are still many problems that need to be solved.Breeding research cannot be delayed.Therefore,in this study,138 colored calla lily resources were used as materials.Based on the correlation analysis of characters,the correlation between traits and EST-SSR markers was analyzed and the EST-SSR associated with the ornamental characters of were identified.In order to provide some theoretical references for the secondary breeding of colored calla lily and its improvement in ornamental quality,the main findings were as follows:1.15 phenotypic traits were observed and simple clustering was performed.Based on this,the correlations between the traits were analyzed.In general,except for special traits,most of the traits had a certain correlation with the rest of the traits providing a reference for analysis of traits and molecular markers.2.Selecting and synthesizing 1,378 pairs of primers that mainly use two-base and three-base repeat motifs for amplification.According to the four DNA pools of BSA,a total of 295 pairs(21%)of the primers had a good polymorphism.The target bands with clear and stable amplification were selected and then 40 pairs of EST-SSR markers were used to analyze the genetic diversity and population structure of 138 calla lily flowers.A total of 125 alleles were detected on 40 pairs of primers,that the heterozygosity was from 0.01 to 0.95 with an average of 0.56 and the PIC values was ranging from 0.15 to 0.75 with the average value of 0.43.These data indicated that the EST-SSR locus obtained in this study were rich in genetic diversity.3.TASSEL software was used to analyze the combination of the above markers with 15 characters including plant height,leaf length,pistil length,phyllosphere length,leaf greenness and so on.A total of 45 related sites were detected under P<0.05 condition.The maximum number of sites associated with phyllopharyngeal tubercle larvae were 10.The number of sites associated with the plant height,the width of the spath,the height of the spathes and the main color of the inner side of the spathes were at least 1.There was a situation in which the same locus was associated with multiple traits.For example,Call Re458 was associated with leaf length,total length of stamens,length of spaths and leaf width.Zacp192 was associated with leaf length,petiole length and total length of stamens and the correlation analysis of phenotypic traits were also expressed.Only 2 related loci were detected under P<0.001 condition and they were related to the trait of fulgurus and throat.
