Preparation Of Ruyanxiao Chinese Medicine Injection And Study On Pharmacodynamics

On the foundation of traditional Chinese medicine powder"Ruyanxiao"prescription and characteristic of Chinese medicine injection prescription, through consulting papers and summarizing many prescriptions of treating the cow mastitis, the prescription of the Ruyanxiao Chinese medicine injection formed from haemolysis test, antibacterial effect experament and compatibility test, witch consists of Flos lonicerae japonicae, Radix scutellariae, Herba taraxaci, Radix et rhizoma glycyrrhizae and Radix angelicae dahuricae.In the studies on the abstraction process, the hypersound abstraction and the traditional water and ethanol method were used to refine the medicine. According to the yield of Chinese medicine extractum, the best ratio of liquid : solid(10:1), abstraction time(60 min) , abstraction number of times(twice) and alcohol sinking number of times (3 times) were confirmed. Some kind of additional reagents are also used in the process of preparing the injection as well as the method of heating the medicine liquid in high temperature and keeping it in refrigeratory. In the end, the colour and clarity steady injection is gained.In the quality control experiment, in accordance with Pharmacopoeia of the People's Republic of China 2005, the protein, tannin and resin are checked. For further quality control, the ultraviolet spectrophotometry method of the chlorogenic acid and the baicalin are studied. In concentration 2.5～10.0 mg/L, the standard of chlorogenic acid curve shows good linearity(R=0.9954, n=6) , its average content is 1.150 mg/mL. The baicalin's standard shows good linearity in concentration 2.0～12.0 mg/L(R=0.9987,n=6), its average content is 0.517 mg/mL.The stability and safety of the preparation has been studied. The result of light stability experiments and heat stability experiments showed that the injection can be preserved two years in the condition of 25℃shady and protection from light.The result of pharmacodynamics studies showed that the injection had determinate depressant effect to the Staphylococcus aureus, Streptococcus mastitidis, Streptococcus dysgalactiae and Bacillus coli. It had a good effect to clinical mastitis (cure rate was 46.67 %, effective rate was 80 %), but the cure rate and efective rate were lower than the penicillin and streptomycin control group(cure rate was 70 %, effective rate was 90 %). The injection had an excellent efect to the subclinical mastitis, the cure rate was 80 %, while the effective rate was 100 %.Through trial and try, a stable, safe and effective Ruyanxiao injection had been preparated. It provided a good attempt for the development and practice of the Chinese medicine injection in animal husbandry.
