In order to develop a kind of veterinary medicine for dairy cattle with mastitis, four compound medicines were prescribed. Compound 1 and Compound 2 were Chinese Traditional Medicines, Compound 3 and Compound 4 were Chinese Traditional Medicines combined with prednisolone. The optimum extraction conditions of Compound Chinese Traditional Medicines were studied by orthogonal test including 3 factors: water-adding amount (A), decocting frequency (B), decocting time (C), and selected by the content of chlorogenic acid (CHA) from dandelion and flos lonicerae. Improved Karber method was used to study the toxic effect of these prescriptions, used LD50 as the evaluate index of the acute toxicity strength. The methods of rats ear swell induced by xylene, leukoplania of rats aerocyst induced by carboxymethyl cellulose and granulation tissue hyperplasia of rats induced by tampons were adopted to study the anti-inflammatory action. The agar proliferation method and tube dilution method were adopted to determine the minimal inhibitory concentration (MIC).The effects of Compound Chinese Traditional Medicines on the permeability of rats cutaneous blood vessel were tested through measuring stained skin area and absorption spectrometry after injecting Evan's blue intravenously. Choosed the best compound medicine basing on all the tests above and prepared Gel used CMC-Na, glycerin and some other materies. Ultraviolet spectrophotometry and rotating basket method were choosed to determine drug-loading and release of Chinese Traditional Medicine Gel. Contents of CHA in the Gel were determinated by ultraviolet spectrophotometry. Stability and irritant of Gel were investigated. Standards for quality control of the preparation were established.The results show that the best optimum extraction conditions of Compound 1 were adding 10 times water, decocting 3 frequencies and 1.5 hours per frequency; the best optimum extraction conditions of Compound 2 were adding 6 times water, decocting 3 frequencies and 1 hour per frequency. The LD50 of compound 1 was 43g/kg, the confidence interval of LD50 was 39~47g/kg; The LD50 of compound 2 was 147g/kg, the confidence interval of LD50 was 133~162g/kg. In the test of inhibiting the swelling of rats ear caused by xylol, the inhibition rates of compound 1, compound 2, compound 3 and compound 4 to rats ear swell by xylene were 67.33%, 63.67%, 70.65% and 71.40% at 24h, 50.50%, 50.55%, 57.59% and 53.51% at 48h respectively. In the test of rats aerocyst leukoplania induced by carboxymethyl cellulose, the leukoplania amounts of the test groups were lower than the control groups, the differences were significant remarkable(p<0.01), The effect of inhibiting leukoplania of compound 4 was the best one among all the four compounds.In the test of inhibiting the hyperplasia of the rats granulation tissue induced by cotton balls, the hyperplasia inhibition rates of compound 1, compound 2, compound 3 and compound 4 were 50.85%, 45.30%, 65.10% and 60.09% respectively. The results of agar proliferation method showed that Staphylococcus aureus was extreme sensitive to compound 2 and compound 4, E. colis and Staphylococcus aureus were midrange sensitive to compound 1 and compound 3.The results of tube dilution method were similar. In the test of inhibiting the raise of vascular permeability of rats caused by xylol, the effect of compound 4 close to prednisolone, was better than compound 1, compound 2 and compound 3. The prescription for the preparation of gel was compound 2, CMC-Na, potassium sorbate, glycerin, sodium sulfite, EDTA and some triethanolamine.The drug loading of Chinese Traditional Medicine Gel was 21.52% tested by ultraviolet spectrophotometry. The equation of cumulative release curve was y=2.9239Ln(x)+3.258. The quality control standards of Chinese Traditional Medicine Gel were that external appearance was henna, texture was uniformity, the content of chlorogenic acid (CHA) was higher than 19.3μg/mL, and the Gel was stable at 60℃and -15℃.In a word, the Chinese Traditional Medicine Gel prescribed for dairy cattles with mastitis had obvious anti-inflammatory and antiphlogosis effect. The preparation of the Compound Chinese Traditional Medicine Gel was simple, the method was accurate, and the standard of the quality control was reasonable.
