| Due to the management level and source purification and other reasons,the poultry breeding industry in our country has complicated diseases.In order to improve the brooding effect,various antibiotics are often used in large quantities.Affecting the green development of animal husbandry,it also directly endangers the safety of human public health.In order to adapt to the development of the trend,adapt to the changes of the times,reduce the use of antibiotics,and find suitable substitutes,this experiment is based on the traditional Chinese medicine brooding experience powder formula.Optimized,developed Qiqing Zhixie Granules,and studied its preparation process,quality control and preliminary pharmacodynamics,which laid the foundation for future clinical expansion trials.1.Optimization of the preparation process of Qiqing Zhixie Granules.The influence of each factor on the extraction of Qiqing Zhixie Granules is C(extraction times)>B(extraction time)>A(water addition);when the ratio of dextrin:sucrose is 7:3,the comprehensive evaluation score is the highest;The forming rate of granules increased with the increase of ethanol concentration.Determine the best extraction process: add 10 times the amount of water,decoct twice for 1.5 h each time,combine the two decoctions,then filter the decoction,make dry extract through rotary evaporation and reduced pressure vacuum drying,grind it.Powder,add dextrin: sucrose ratio of 7:3 auxiliary materials,use 95% ethanol as a wetting agent,spray the mixed powder,dry,the effect of preparing granules is the best,and the process is stable and reliable.2.Study on the quality control of Qiqing Zhixie Granules.The 4 batches of Qiqing Zhixie Granules all meet the relevant provisions of Part II of the 2020 Edition of the Veterinary Pharmacopoeia of the People’s Republic of China;the 4 batches of Qiqing Zhixie Granules and the baicalin reference substance all show the same color on the corresponding chromatographic position.spot.The determination results showed that the regression formula of baicalin was y=0.073x-0.0255(R2=0.997),and the linear relationship was good in the concentration range of 2-10 μg/mL.The average recovery rate was 98.18%,and the RSD value was 1.66%.method is stable.The method is suitable for the content determination of baicalin,the active ingredient of the monarch medicine of Qiqingzhixie Granules,and the measured active ingredient of baicalin is not less than 4.00%.3.In vitro and in vivo antibacterial test of Qiqing Zhixie Granules.Qiqing Zhixie Granules have a certain bacteriostatic effect on a variety of avian pathogens,can effectively improve the survival rate of chicks and alleviate the weight loss caused by pullorum infection,and have a certain growth-promoting effect.4.Toxicological study of Qiqing Zhixie Granules.The maximum tolerated dose of Qiqing Zhixie Granules is 180.0 g/kg,which has met the safety standard.The subchronic toxicity test showed that 28 days of continuous feeding had no significant effect on the growth and development of rats,and the drug had no toxic side effects and was highly safe.Conclusion: The preparation process of Qiqing Zhixie Granules in this study is stable and feasible,with good repeatability,accurate and reliable quality control method,good bacteriostatic effect and certain growth-promoting effect,and the drug has no toxic side effects and high safety. |
