Wheat male sterile line with Ae.kotschyi cytoplasm keeps itself in the foreground compared with others,and the effect analysis of fertility restorer genes is helpful to select good restorer lines and hybrid wheat with high heterosis.Using the sterile line Yumai 3,the corresponding maintainer line,and the restoring line Yumai 2, the F2 population Yumai 3/Yumai 2 and the backcross populations K-Yumai 3 / F2 plants were crossed to study the effect of the two restorer genes by means of the SSR makers and fertility segregation ratio.The results were as follows:1)The genotype of each plant for the F2 population was estimated by means of the SSR markers linked with the restorer genes and fertility segregation ratio of the backcrosses.In the F2 population,there were 16 plants with the genetype Rf1Rf1Rf2Rf2,21 plants with the genetype Rf1Rf1rf2rf2,12 plants with the genetype rf1rf1rf2rf2,32 plants with the genetype Rf1Rf1Rf2rf2,37 plants with the genetype Rf1rf1Rf2Rf2,40 plants with the genetype Rf1rf1rf2rf2,23 plants with the genetype rf1rf1Rf2rf2,and 76 plants with the genetype Rf1rf1Rf2rf2.The segregation ratio of the 9 genotypes was fitted to the ratio 1:1:1:1:2:2:2:2:4(P>0.05).2)On only the Rfl gene was existed,the average restoring degree was 68.49%for Chinese method and 103.71%for International method under the dry condition,and that was 60.90%for Chinese method and 94.16%for International method under the normal condition.3)On only the Rf2 gene was existed,the average restoring degree was 54.39%for Chinese method and 83.38%for International method under the dry condition,and that was 50.56%for Chinese method and 78.22%for International method under the normal condition.4)On both of the genes were existed,the average restoring degree was 82.33%for Chinese method and 126.24%for International method under the dry condition,and that was 79.31%for Chinese method and 121.32%for International method under the normal condition.5)The effects of the two restorer genes were different,The effect with the restorer gene Rf1 was higher than that with the restorer gene Rf2 under the two conditions. The effect with the two genes was higher than that with only one gene,and the two restorer genes acted in an additive effect.
