Tetracycline was widely used as antibacterial to pigs, cattle, chickens, ducks and other animals in the domestic and international. And it often used as additive add to feed to prevent diseases. Because of extensive application for a long time, it engendered drug-resistant bacteria, the use of dose rising unceasingly. Also, because of high-dose abuse for a long time, in animal tissue tetracycline residues rised. When people eated these food, it could bring greatly harm to health. It already both at home and abroad reported that it could trigger "double infection", tetracycline teeth, foetus' death. It also could be teratogenic and carcinogenic for a long time use.This study was designed to 4-week-old chickens and 15-day-old ducks for an accumulation of tetracycline toxicity experimentation. this test adapted single level three dimensional design, the test animals were divided into four treatment, group 0(comparison group), groupⅠ(low dose), GroupⅡ(midding dose) and groupⅢ(high dose). According to the design, giving each dose group each chicken and duck oral dosage 100mg/(kg.d), 200mg/(kg.d), 1000mg/(kg.d). This experimentation tested animals of different doses of oral tetracycline performance, production performance, clinical symptoms after animal take orally different dosage of tracycline, cut open and examine changing, histology slices, the biochemical index of blood plasma and measuring, tetracycline remains in tissue with the modern most advanced instrument high-efficient liquid phase chromatograph (HPLC). It could offer a scientific basis for the production of timely detection of tetracycline substandard animal products and prevention early tetracycline residues exceeding.Chicken accumulating toxicity test results①symptoms before death: after taking tetracycline, chickens in the test group had appeared poor in spirit successively, the appetite decreasing, addicted to sleeping, fluffy feathers, the appetite is reduced by half or abolished definitely, does not get up lying, eyes half-closed or close all, cockscomb and face pale,watery shit.②Anatomy changes: Cut open and examine the natural death chicken and chicken in one month timly, liver and kidney obviously intumesced, the size was as big 1~2 times as comparison group liver. The Diver marginal was obtuse, there were hemorrhagic and necrotic lesions, kidney intumesced, bleeding red. Pericardial effusion and swelling.③Biopsy: Regard liver change necrosis as main characteristic, liver little leaf cell arrange very much disorderly. Cardiac muscle became enlargement, the gap widened. Small intestine floss became shedding necrosis and behind floss there were Flock inflammatory cells.④Plasma biochemical index: Reaction liver, kidney and heart damaged in the blood biochemical indicators increased. Early detection of cancer and biochemical indicators of a fetal antigen (AFP) and carcino embryonic antigen (CEA) were negative.⑤Detection of chicken tissue residues: muscle group 0,Ⅰ,Ⅱ,Ⅲrespectively ND,0.14,0.39.1.33 (mg/kg). Chicken Liver: Group0,Ⅰ,ⅡandⅢrespectively ND,0.25,0.47,1.63 (mg/kg). Noted that groupⅠ,Ⅱ,Ⅲmuscle and liver tetracycline residues volume exceed NY5034-2005 standards.Duck's accumulating toxicity is tested. Tetracycline residues repeated experiments: symptoms before death ducks were more mild than chickens, and no one death. After dissection, I found that liver significantly enlarged, yellowish-brown, some forms bleed in order to mainly behave. Histological slices in the liver, kidney necrosis. Plasma biochemical indicators noted that the liver, kidney, heart damaged mainly. HPLC detection of residues of tetracycline,Ⅰ,Ⅱ,Ⅲmuscle and liver were high, exceed NY5034-2001 standards.The innovation: This test creatively did chicken tetracycline accumulation toxicity test. studied the animals' symptoms before death, cut open and examine changing, Histological sliced, plasma biochemical index and HPLC detection of residues in tissues. While take meat duck as repeated experiments, the meat duck's test result keeps the same with chicken. These results reflect the scientific integrity of the tetracycline toxicity, absorption, distribution, degradation and residues testing. The research content were rarely reported. It was advanced and innovative. It had important production value and practical significance.
