Using the method of"random sampling in typical colonies of the central area of the habitat"and seven microsatellite markers, we examined the genetic variations of 70 Bayinbuluk sheep and estimated their gene frequencies . The same data of Hu sheep , Tong sheep, Xiaowei Han sheep, Tan sheep and Ujumuqin sheep ,which served as the referenced population, were cited to further explore leves of genetic differentiation. The mean heterozygosity (H) of Bayinbuluk sheep was 0.8208 ; It's average polymorphic information content(PIC) values was 0.8091;The mean effective number of alleles(Ne) valued 6.9550 . The values of these three parameters showed the degree of genetic variation in Bayinbuluk sheep in non-coding regions was highest than other sheep populations .The coefficient of gene differentiation among five sheep populations was 0.026254 , and that among six populations was 0.039272,indicating that the degree of gene differentiation was comparatively low among sheep populations .But the coefficient of gene differentiation among six sheep populations was higher than five sheep populations, indicating that the gene differentiation among Bayinbuluk sheep and other five sheep populations was high. Hu and Tong sheep were grouped at frist , Tan and Ujumuqin sheep clusted , and then clusted with small Tailed Han sheep , finally Bayinbuluk sheep clusted with all above. The relationships between sheep populations were closely related to the distances between sheep populations.The genetic diversity for 11 encoding blood proteins structural loci were studied using polyacrylamide gel electrophoresis (PAGE) and starch gel electrophoresis (SGE) in Bayinbuluk sheep, and compared with other 4 sheep populations among Hu sheep , Tong sheep, Xiaowei Han sheep and Tan sheep in China. The average heterozygosity of Bayinbuluk sheep was 0.3514,It's average polymorphic information content was 0.2900, It's average effective number of alleles valued 1.7832 , It showed the degree of genetic variation in Bayinbuluk sheep in coding regions was highest than other sheep populations . The coefficient of gene differentiation among four sheep populations was 0.0868. and which among five populations was 0.1207, indicating that the degree of gene differentiation was comparatively low among sheep populations , as the same as the result of the microsatellite marker . The genetic relationship between small Tailed Han and Tan sheep was closest ,Hu and Tong sheep, which between followed, and Bayinbuluk sheep was the farthest from the above.The relationships between sheep populations were closely related to the distances between sheep populations.The gene frequencies of 7 microsatellite loci and 11 structural loci in Hu , Tong, Xiaowei Han ,Tan and Bayinbuluk sheep ,which were sampled by he method of"random sampling in typical colonies of the central area of the habitat", were comparatively analyzed and discussed to verify the relationships of genetic differentiation among populations . The results showed the heterozygosity, polymorphic information content and effective number of alleles at each microsatellite locus and their average were higher than those at each structural locus , which indicated that the levels of population variations revealed by the microsatellite loci were higher than those on the structural loci .The coefficients of gene differentiation computed by the data of gene frequencies at the microsatellite loci were lower than those at the structural loci . The levels of gene differentiation among populations were low on the whole .The standard genetic distances among populations based on the microsatellite loci were higher than the structural loci .The relationships among populations displayed by the trees constructed from DS distances according to the either data of gene frequencies at the microsatellite loci or the structural loci were not consistent. The dissimilarity required further verification .
