Applying the method of simple random sampling in typical colony, 63 Hu sheep and 65 Tong sheep were sampled in their central area of habitat Huzhou city zhejing privince and Baishui county Shanxi province respectively. Each individuals had independent blood line. The body measurements were surveyed and the phenotype frequency related to morphological characters were observed. Starch gel electrophoresis was used to detect the variations of 14 blood enzyme and other protein structural loci, and the reliability and accuracy of every estimated frequencies value were evaluated. The polymorphisms of 7 microsattllite DNA loci distributed on 6 different chromosome were examined by combination of PCR amplifying and 8% denaturing polyacylamide gel electrophoresis (PAGE). The related data of 15 sheep populations home and abroad was quoted. At same time 49 Yangtse River Delta White Goats (Cbs) were sampled as contrast population from the suburb of Yangzhou city Jiangsu province by the simple random sampling method. The results were as follows:1. Hierarchical Clustering of 6 sheep populations based on 17 quantity indexes including body measurements, ecological and morphological characters indicates that 6 sheep populations can be classified in two types: the first includes Mongolia sheep (Moy) and Tan sheep (Tan) distributed in husbandry area and the juncture of agriculture and husbandry area, the second includes Daweihan sheep (Dwh),. Xiaoweihan sheep (Xwh), Tong sheep (Tong), Hu sheep (Hu) distributed in agriculture area. The Principal Component Analysis of 17 quantity indexes are carried out. R-type Hierarchical Clustering based on the principal component of each population produces results resembled to the former. The 17quantity indexes about population ecological and morphological characters are studied with Q-type Hierarchical Clustering method: the elevation and mean annual rainfall are found to be the important factors influencing breeds classification. So it is difficulty to discriminate the phylogenetic relationship among populations based on information about ecology and morphology, but the data about ecological and morphological characters can be used as the important auxiliary material for the research of phytogeny status.2. 14 structural loci and 40 alleles in blood enzyme and other protein variations of Hu sheep and Tong sheep populations were examined. After collecting the data of 10 same loci about 15 sheep colonies home and abroad, mean Heterozygosity of each population was compared, matrix of fuzzy similarity relation describing the genetic similarities of different population was established and composed. Results show that. There are high Heterozygosity for Hu sheep, Tong sheep and Mongolia populations and low Heterozgosityfor European populations. The South Asia populations lie between the two formers. There are closely phylogenetic relationship among Hu sheep, Tong sheep and Mongolia populations. Mongolia sheep populations have relatively independent phylogeny status compared with the South Asia and European sheep populations.3. Collecting the data of Ke locus about 11 sheep populations home and abroad, the genetic variation of Ke locus was analyzed. Using "dominant-dominant" and "dominant-co-dominant" models, the genetic disequilibrium between Ke and other 12 structural loci was examined. The results indicates that: Under the condition of abominable ecology and climate, there is evident superiority to select HK and Keh, There is relativity between Ke and Tf (p<0.01) in the model of "dominant-co- dominant ", Tf locus is in Hardy-Weinberg disequilibrium. The reason of genetic disequilibrium needs further study.4. 7 sheep microsatellite markers (OarFCBll, OarFCB128, MFA70, OarAElOl, MAF33, OarFCB48 and OarFCB304)located on chromosome 2, 4, 6, 9, 17, 19 respectively were selected for examine the samples of Hu sheep, Tong sheep and Yangtse River Delta White Goat (Csb) through PCR amplifying.
