Cordyceps militaris as a medicinal fungus, has a long application history in China. In recent years series of studies showed that, C. militaris has various biological activities, in particular inhibitted such as tumor and virus. C. militaris BYB-08 obtained by protoplast fusion previously was studied in food-grade fermentation with the targets of optimizing crafts and obtaining stable culture. Finally, benifical and instant food main in culture by C.militaris BYB-08 were exploitatied out.The important results in study include:1. The method of moving average was used to find the end of liquid fermentation, and the results showed that:during the time of 108~114h, the liquid culture was the best as seed, because of fastest mycelial growth, and not less biomass production.2. In solid fermentation, all materials were food-grade. Tests were conducted in main selection by comparison, elementary selection by uniform design and precise selection by Box-Behnken design. After that, the best compounding of solid culture material was determined:wheat, rice, oats, yeast extract (1:1.02:1.12:0.20). Finally, C. militaris mycelia growth rate compared with 38.6% increased before optimization.3. Soaking, pre-gelatinizing, rapid cooling, and decentralization liquid addition were used as the improved technologis for characteristics of fermentation material, witch made decentralization of materials reach 90% or more. After optimizations, mycelia could cover with the grain surface in 4days.4. Taking optimized crafts, fermentation products had consistent quality and low contamination rates. Analysis of solid fermentation material by C.militaris BYB-08 showed, the total number of colonies was 625cfu/g, which far below the national standard 5×104cfu/g. With 4 batches and a total of 20 tests, the results showed that polysaccharides production was 1.04±0.012mg/g, and cordycepin production was 1.87±0.024 mg/g.5. Two kinds of convenience food and a kind of instant drink product main in solid culture of C.militaris BYB-08 were empoldered. The sensory evaluation tests of these products were finished, Then, testing of product qualities were carried out by Centers for Disease and Health Control of Hubei Province, and the results showed that, products were qualified. Finally, crafts was established, and quality standards was set for the food producst reference to national standards.
