The Aphis is a destructive vermin of wheat,which makes wheat yield lose and quality low throughout the world.Breeding and popularizing resistance cultivars has been proved the most effective,economical and environmental safe method to suppress the midge broad-spectrum Aphis resistance.Therefore the exploitation of insect-resistant genes,including its marking,location and molecular-marke aided selection,would benefit for the breeding effective of the Aphis resistant cultivars.In this study,the resistant gene of Aphis of wheat Zhengkel was analysis by using SSR technique.The results are showed just as follows:1.The 495 of F2 individuals and their Zhengke l(resistant mother)and Shi4185 (susceptible father)carrying unknowing gene were inoculated with Aphis strain at seedling stage to assess their resistant ability.Genetic analysis suggested that the resistance of Zhengkel was controlled by a pair of single dominant gene.2.Ten of 495 DNA samples extracted from the F2 population were pooled into two separate groups for bulked segregant analysis(BSA).By SSR analysis with 3 pairs of primers located on 7D chromosome to the insect-resistant gene of Aphis which round Xgwm437,Xgwm44,Wms350.3.Three markers located on the 7D chromosome to the Zhengkel insect-resistant gene of Aphis.We screened a molecular marker Xgwm437 and Xgwm44 that linked to the Zhengkel resistant gene,the genetic distance among marker Xgwm437,Xgwm44 and the resistance gene was calculated as 10.49cM and 3.29cM.Xgwm44 was regarded as an assistant selecting marker for gene.4.Compare with last study,the gene was denominated DnY transitorily,which be located on 7D chromosome.
