In this experiment were obtained through 311 species of wild edible fungi,including 14 species of Ascomycetes and 297 species of Basidiomycetes.We identification of the reliability and classified status of these species of strains,and finally 254 strains isolated success and were classification.The molecular method can identify species of fungi using mycelium.In past study, RAPD,SRAP and ISSR were major techniques applying to molecular identification of edible fungi mycelium.However,those techniques are less reliable and lack enough accuracy.In this study,species identification of wild edible fungi mycelium was conducted based on DNA sequences in the ITS region of rDNA.The method writes as follows:ITS regions of rDNA of fungi identified are amplified and sequenced,the sequences determined were compared with those registered in DNA sequence database on Internet,then identification results were obtained according to comparing results and other biological information in DNA sequence database on Internet.The final classification results were bassed on Morphological identification result,Molecular identification result and Characteristics of strains.In 14 species of Ascomycetes,Pollution is 122,143,140,391,141,128,LJ0602,LJ0601,389,142,QMY005,the final Accessed 3 strains.From molecular identification Correct name 2.In 297 species of Basidiomycetes,in accordance with sub-divided,Divided into four categories for analysis.At the Heterobasidiomycetes:In 24 species of Phragmobasidiomycetidae,Pollution is 235,0002,From molecular identification Correct name 13,the final accessed 22 strains.In 133 species of Holobasidiomycetidae,Pollution is XS006,QMY014,070,146,078,TS0004,065,0019,068,0027,0020,208,355,198,From molecular identification Correct name 60,the final accessed 117 strains.At the Hymenomycetes:In 134species of Holobasidiomycetidae,Pollution is C08,C33,TS0006,C78,QMY038,TS0005,C24,L030,C14,LMS012,ZWY018,QMY035,ZWY003, C02,XS024,HS041,C37,0010,QMY012,ZWY002,XS043,C01,QMY032,HS072,008,074,From molecular identification Correct name 12,the final accessed 108 strains.In 8 species of Gasteromycetidae,Pollution is WP黄,HS020,XS047,TS0002,From molecular identification Correct name 4,the final accessed 4 strains.During research,I discovered:1,In the identification of strains,alone in the use of Conventional identification methods was difficult,and prone to errors,the combination of molecular identification techniques can be applied to the identification of srains is more convenient,more accurate results.2,Molecular identification techniques for the identification of wild mushroom strains is on the basis of the molecular level,but there are questions should be combining with the other techniques such as morphological character,is easier to identify disputed strains.
