This experiment mainly through compares butorphanol compounded propofol as induction of anaesthesia to propofol as induction of anaesthesia only,the effects of.cardiorespiratory function and,clinical anesthesia and pharmacokinetics of propofol by constant speed administration were investigated, discusses the feasibility of combined anesthesia with butorphanol and propofol,will settle the foundation for application of propofol anaesthesia small dogs in the future.The dogs were divided in two groups.Group one was performed induction of anesthesia with 4 mg/kg of propofol ,and group two was performed induction of anesthesia with 4 mg/kg of propofol compounded 0.2mg/kg of butorphanol.Both of the two groups progressed maintenance of anesthesia with 0.4 mg/(kg min) of propofol. Measured HR,SpO2,MAP, PR,T by Datex invigilator. ,and evaluate the effect of sedative,analgesia,muscle relaxation,kinds of physio- reflex after anaesthing The different time plasma drug concentration of propofol was monitored by HPLC- fluorescence detection.The result indicated that anesthesia with butorphanol-propofol complex induction was completed in 90s.HR rose up after 2min and fell-off to the normal gradually. Effect of sedative and analgesia can maintain 8-10min.During the time of anesthesia,the T fell,MAP and RP rose up,but they allwithin normal limits. SpO2 was sustained around 90%.The dogs waked up after 13min. Induction of anesthesia was fast, stable,and no adverse effect.Two groups were performed induction of anesthesia with 4 mg/kg of propofol and 4 mg/kg of propofol compounded 0.2mg/kg of butorphanol respectively, and all progressed maintenance of anesthesia with 0.4 mg/(kg min) of propofol. Monitored the dogs by clinical evaluation and Datex invigilator, then analyzed and contrasted the results. It indicated that the group of propofol can maintain anaesthing for 70min, mitigating for 68min, analgesia and muscle relaxing for 35min, hut the group of butorphanol-propofol can maintain anaesthing for 72min, mitigating for 69min, analgesia and muscle relaxing for 45min. During the maintenance of anesthesia, the value of T fell a little, HR and MAP increased midrange, SpO2 maintained about 90%, RR in the group of propofol increased slightly, but in the group of butorphanol-propofol decreased slightly. There is no significant deviation between the change of index in two groups before and after anesthesia, also no significant deviation between two groups. The result indicated that butorphanol- propofol combined induction can increase the effect of propofol anesthesia,and do not effect the physiological function. Dogs were performed maintenance of anesthesia with 0.4 mg/kg of propofol intravenously (IV) by the microinfsion pump, and plasma was collected and anticoagulated with heparin at 2,5,10,15,20,30,45,60 and 90min after the beginning of the propofol infusion respectively. The extractive of propofol was redissolved in high-performance liquid chromatograph (HPLC) mobil phase and was detected by the combination of HPLC and fluorescence detection. The results of pharmacokinetics indicated that metabolism of propofol has been identified as the appropriate model for pharmacokinetic analysis of propofol disposition in dogs. The pharmacokinetic parameter as following, distribution half-life (T1/2α) was 4-11 min, elimination half-life (T1/2β) was about 28 min, body learance (ClB ) was 5-6 L/kg/h, elimination rate constant (Kel) was 4.36±1.15h, and apparent volume of distribution (Vd) was 3.31±1.14L/kgt. In the group of propofol,firstly,the plasma drug concentration rose rapidly,then the value maintain around 13μg/ml after 20-45min of anesthesia.60min later,the value of plasma drug concentration fell to 5.4μg/ml.90min later , the value of plasma drug concentration fell to 1.9μg/ml.In the group of propofol compounded butorphanol,firstly,the plasma drug concentration rose rapidly.The value reach the top 14.1μg/ml after 30min,then fell .It fell to 6.5μg/ml after 60min ,and to 2.5μg/ml after 90min.there were no obviously difference in plasma drug concentration of propofol with the change of time administer,which indicated that small dosage butorphanol has no obviously affect on propofol target controlled infusion anaesthesia.An experiments was progressed in order to authenticate the effect of butorphanol-propofol combined anesthesiain surgery anesthetic. The results showed that dogs were narcotic rapidly,and did not respond to discissio the skin,blunt dissection the muscle,drag the intestine and stomach and so on operation. There was no adverse effect so the surgery progressed smoothly.The dogs came round soon after the finish of the operations and injection,and the dogs hads no narcoma. Effectiveness of anaesthesia retain 45-60min, can consistent with the requirement of anaesthesia used in common and diminutive operation.
