With its simplicity, exogenous total DNA introduced via pollen tube pathway has been widely applied in China and new varieties or strains have been obtained in more than 20 kinds of crops. The sweet sorghum has the properties of drought-tolerance, waterlogging-tolerance, infertility-tolerance, alkalic salt-tolerance and so on, whose biological output, sugar content and the conversion rate of ethyl alcohol is high. It belongs to fine grass crop, sweetener crop and renewable energy crop. The paper introduced total DNA of sweet sorghum into maize inbred line via pollen tube pathway. Combined with molecular markers and field investigation, Variation was screened so as to Widen maize germplasm resources and establish the technology of rapid identification of exogenous DNA introduction maize variant offsring. The main results were as follows:(1)The total DNA of sorghum was introduced into maize inbred lines 4112 and 8902 via pollen tube pathway technique. Based on the results of stem sugar content and molecular testing, 17 Variants for D2 generations was screened.(2)10 random RAPD primers had polymorphism among donors, receptors and offspring. The amplified fragments include three types: non-biased-parental type, biased-donor type and biased-receptor type. The results provide some direct evidences of molecular biology for molecular confirmation.(3)The results of field investigation for 17 Variants showed that it had extremely obvious difference on the axis diameter between 4112 and its offsprings. The results also showed that it had extremely obvious difference on the ear length and axis diameter and had obvious difference on the ear weight and kernel weight between 8902 and its offsprings, respectively.(4)The results of SSR marker were consistent with those of RAPD marker.(5)The specific fragments by RAPD marker were recovered and sequenced. Based on the sequencing results, specific primers of SCAR marker were designed and applied in PCR testing so as to confirm the effectiveness of specific primers.
