Agaricus Blazei Murill Degradates Arabinoxylan And Improves The Nutrition Quality Of Wheat Bran

In this series of three parts of the experiments,we analyzed the contents in the mixture of liquid fermentation;evaluated the potential biological activity,including advancing the functional structure of intestine,promoting the digestion function and enhancing the immunity in mice.In these experiments,wheat bran or corn meal were used as the main carbon resource of Agaricus blazei Murill(A.b)and the total content of the fermentation system were used as an additional part of the components in daily diets.Our result shows that A.b can undermine the anti-nutrition activity of arabinoxylan in wheat bran by its unique biological degradation activity.ExperimentⅠ,the concentration of arabinoxylan,total sugar and protein in different liquid mediums followed different cultural protocol was compared to evaluate the degradation ability of A.b to wheat bran.In the groups of no-filtering protocol(AW & AWC),the concentration of these metabolism production is much higher than those in the groups followed filtering protocol(AWCL & ACL)(P<0.05).In addition,the inoculated group(AW) contains much more sugar and protein than the no-inoculated group(WB)(P<0.05),though the concentration of arabinoxylan in these two group is similar.(P>0.05)these results indicated that A.b xylanolytic enzyme system can co-degrade cell wall polysaccharides and nitrogen nutrition inlayed in it,this biological activity can greatly increase the concentration of arabinoxylan,total sugar and nitrogen nutrition in fermentation liquid.ExperimentⅡ,polysaccharides were extracted from fermentation liquid of three different group,group of wheat bran and corn meal(AWC),group of filtered wheat bran and corn meal (AWCL),and group of filtered corn meal(ACL),and the monosaccharide were identified. The result shows that,the main polysaccharide contents from fermentation liquid of AWC and AWCL were occupied by D-Glc,L-Ara and D-Xyl,while the main monosaccharide contents from ACL fermentation liquid was D-Glc.What's more,arabinose and xylose were the main degradation production of wheat bran arabinoxylan,while glucose was main hydrolysate of polysaccharide from corn and wheat bran,and the hemicellulose components degraded by A.b can also release glucose.This result can match with that of experimentⅠwell, so we can draw the conclusion that A.b can effectively degrading and transferring structural polysaccharide of wheat bran.ExperimentⅢ,mice were fed with four different diets,conventional diet(C-CK), mixture of conventional diet and the total content of wheat bran-A.b fermenting production(C-AW),mixture of purified diet and the total content of wheat bran-A.b fermenting production(P-AW)and mixture of purified diet and un-treated wheat bran(P-CK). The change of body weight,digestion ability and immunity were observed to evaluate the reducing effect of A.b degradation to the anti-nutritive effects of arabinoxylan.The potential enhancing ability of nutrition immunity of these metabolic products is also observed in this experiment.The result shows that,increasing of weight and feed-weight ratio of mice in group P-AW were significantly superior than group P-CK(P<0.05);while the average length of intestinal villous was significantly increased than the group P-CK(P<0.05),crypt depth significantly decreased(P<0.05),and VCR significantly increased(P<0.05),and the D-xylose concentration in serum of mice was higher(P<0.05)of mice in group P-AW.In addition,spleen index and thymus index of the mice in group P-AW and group C-AW was higher(P<0.05)than those of the control groups(P-CK,C-CK)respectively;the antibody level of Bovine serum albumin(IgG)from the mice in group P-AW was significantly enhanced(P<0.05)than those from untreated wheat bran groups.These results indicated that the A.b fermenting products of wheat bran can improve not only digestion-absorption of small intestine but also specific immunity of the body.Nevertheless,the further study would be required to evaluate its effect in animals.To sum up:A.b can effectively degrade wheat bran by its unique extracellular enzyme system;the degradation activity can reduce the anti-nutritive effects of arabinoxylan in wheat bran,release the nutrition inlayed in the cell wall,and thus increase the nutrition quality of wheat bran.The total content of A.b fermentation of wheat bran can improve both the absorptive capacity and immunity in mice;this biological ability is far stronger than that of the untreated wheat bran and corn meal.
