With the development of society,the application of heavy metals is more and more extensive,but the phenomenon of heavy metal poisoning is gradually increased.This experiment mainly discusses the protective effect of Agaricus blazei polysaccharide on cadmium-induced injury of chicken testis,On the application of the theoretical basis.In this study,80 7-day-old broilers were randomly divided into four groups: blank control group(normal full-price feed),polysaccharide group(normal full-price feed,oral 6 mg Agaricus polysaccharide),cadmium group 140 mg / kg Cd Cl2)and polysaccharide plus cadmium group(mixed with 140 mg / kg Cd Cl2 the normal full price of feed,while daily oral 6mg Agaricus polysaccharide).At the 20 th,40d and 60 d,the testis tissues of chickens were used to calculate the testis organ index on the chickens and their testes.The levels of cadmium in the testes,antioxidant enzyme superoxide dismutase(SOD)and glutathione(GSH-Px)activity,malondialdehyde(MDA)content,expression of inflammatory factors(TNF-?,IL-1? and IL-6),detection of apoptosis genes,The testis tissue of 60 d chickens was made.The test results showed that the testicular organ index of polysaccharide plus cadmium group was significantly higher than that of cadmium group at 40 days and 60 days(P<0.01).The content of cadmium in the testes of polysaccharide plus cadmium group was on the 20 th day,40 th day and 60 th day(P<0.01).The activity level of SOD and GSH-Px in testis tissue of polysaccharide plus cadmium group was significantly higher than that of cadmium group at each time point(P<0.01)(P<0.01).The expression of inflammatory cytokines TNF-?,IL-1? and IL-6 in the testes of polysaccharide plus cadmium group was significantly lower than that of cadmium(P<0.01)at 60 days(P<0.01).The expression of Bax and Casp-3 mRNA and protein in the testes of polysaccharide plus cadmium group was significantly lower than that of cadmium group(P<0.01).The expression of Bcl-2 The mRNA and protein levels of the gene were significantly higher than those in the cadmium group(P<0.01).The results showed that cadmium exposure could induce cadmium accumulation in chicken testis,inhibit the activity of antioxidant enzymes(SOD and GSH-Px),increase the content of MDA,cause the expression of inflammatory factors,and accompanied by severe tissue injury and inflammation Sexual infiltration.The Agaricus blazei polysaccharide can reduce the accumulation of cadmium in the chicken testis,significantly increased the activity of cadmium-inhibited antioxidant enzymes,decreased the content of MDA and the expression of inflammatory factors,thus attenuated oxidative stress and inflammatory response,relieved cadmium on testes The toxic effects of the organization.In this experiment,it was clarified from the level of gene and protein.The testis injury of cadmium could cause testicular injury.After adding Agaricus blazei polysaccharide,cadmium decreased the damage of testis,which indicated that Agaricus blazei polysaccharide had protective effect on cadmium-induced injury.Which provided a theoretical basis for the study of active ingredients of traditional Chinese medicine,and provided a new idea for the prevention and cure of cadmium poisoning.
