As a world famous flower lily is widely application in many aspect. This ex -epriment is based on the material of L.henry(Wudangshan,Shennongjia), Japanese lilium(series of X) and Asiatic hybrid (Siberia, Sorbonne ) ,we carried out on the study of establishment of rapid propagation system, flowering time regulation,exa -mination of pollen vitality, lily crossbreeding and culture of the filial generatio-n. The results were as follows:(1) The best inducing medium for callus was MS + 6-BA0.4mg/L that was including Wudangshan, Siberia, sorbonne. their callus rate was respectively 94.0%,96.4%, 95.8%; The best inducing medium for shennongjia was MS+6-BA0.6mg/L, its callus rate was 83.8%.The best rootage medium for Sorbonne was MS+ NAA1.0mg/L; Rooting rate of Wudangshan, Shennongjia and Siberia was best on the medium of MS+ NAA 0.6mg/L.The best Transplanting medium for Wudangshan was yellow sand: blue sand=2:1,the transplanting survival rate was reaching 91%(2) In the flowering time regulator experiment ,elongating light time treatment for 20d and 30d was very significant higher than 10d treatment. It was not significant between 20d and 30d treatment; the effect of GA3 was not significant, the effect of KT and NAA was very significant.the best hormone combination was NAA300mg/L + KT150 mg/L +100 mg/L GA3 once, it is 4.5d earlier than CK.(3) The best pollen germination medium was 20% sugar + 100mg/kg boric acid. The best temperature for super low temperature store was -71℃. Pollen germination rate was 16%afte 12d.(4) The cut-style pollination results was follows: the cross combination(direct-cross and reciprocal cross) of X5 (♀)×L henry (Wudangshan,Shennongjia) (♂),X9(♀)×L. Henry(Wudangshan,Shennongjia) (♂),X27 (♀)×L .henry(Wudangshan,Shennongjia) (♂) and Sorbonn (♀)×L. henry(Wudangshan,Shennongjia) (♂) was very compatible;the cross combination of Wudangshan (♀)×S5 (♂) have the highest rate of swelling capsule ,it was 57.1%. The cross combination of S8 (♀)×Wudangshan (♂),S12 (♀)×Wudangshan (♂),S20(♀)×Shennongjia (♂) was completely incompatible.(5) The best medium for culture of the filial generation was MS+NAA0.2mg/L+6-BA0.1mg/L with ovary slice culture.it was best with B5+ 40mg/L sugar for embryo culture...
