Salvia miltiorrhiza Bunge is a kind of traditional and rare medicinal herbs, Whichcontains a variety of medical composition and has an attractive development utilizationforeground.The callus was induced and proliferated by using young leaf as explants.The effectsof the callus、the adventitious bud and the root on induction and proliferation werediscussed by using different kinds of hormone and different genotypes.The growth circle and some physiological and biochemical factors were studiedduring the culture of callus,and a set of rapid,effieient and steady propagation techniqu-es were founded.All data we got in this paper further provided scientific basis for tissue culture andsecondary metabolism matters yield, and provided technical support for application ofrapid propagation to practical production.In addition, the genetic variability has been analyzed between eight generationsfrom the vegetative propagation, including the morphological characteristics and thegenome level.The results are listed as following:1. Induction of callusDifferent concentration of hormone and genotypes were found to have differenteffects on induction and proliferation.Medium MS+6-BA1.0mg/L+NAA1.0mg/L wasthe optimum medium for callus induction.The fourth was the best for the induction ofthe callus.2. Induction and proliferation of tufted budMedium MS+6-BA1.0mg/L+NAA0.01mg/L was the optimum medium for callus i-nduction, and the fourth was the best for the induction of the tufted bud.3. Adventitious rootsThis study showed that the optimum medium for callus induction was the medium1/2MS, where the roots grow, are earlier, more, healthier and stronger.4. Study on physiological and biochemical charactersThe growth circle of caihs was28days and the growth curves looked1ike“S”under the condition cultured in1ight and dark.During the growth circle, the tendency ofsoluble protein contents, SOD actives and POD actives were different during thegrowing.They showed some physiology and biochemical characters during callusproliferation and cell actives.The growth curve of callus cultured was different from inbright and that cultured in dark.5. Study on the genetic variability of regenerated plantsNo mutation was observed for different regenerated plants to leaf size, leaf colour,plant type and the genome level between the eight genominational lines....
