Genetic differences in 4 strains of Lentinula edodes and 4 strains of Pleurotus(two belonging to Pleurotus ostreatus,one belonging to Pleurotus nebrodensis,the other belonging to Pleurotus citrinopileatus)were examined using AFLP and SRAP.For AFLP, the genetic similarity coefficient between the four strains of L.edodes,the two strains of P.ostreatus,the three species of Pleurotus as well as Lentinula and Pleurotus was 0.74~0.89,0.89,0.47~0.89 and 0.44 respectively.And for SRAP,the relevant coefficient was 0.70~0.93,0.93,0.47~0.93 and 0.53 respectively.So we can infer that the genetic similarity coefficient between strains is above 0.70 in the same species;0.5~0.7 between P.ostreatus,P.nebrodensis and P.citrinopileatus;not less than 0.5 between Pleurotus and Lentinula.The results of two kinds of molecular marker analysis showed that the full sib and half-sib of two strains of Pleurotus ostreatus was classified precedently into a group exhibiting things of one kind come together respectively,that is,they are,and the genetic similarity coefficient of half-sibs was less than that of full sibs.In addition, antagonistic response intensity of half-sibs was stronger than that of full sib,indicating consistency with the results of molecular marker analysis.The cluster analysis result of the full sib and half-sib ofLentinula edodes also showed that the tested strains of the full sibs and half-sibs were classified precedently into a group respectively by using SRAP, compared with the useing AFLP,the trend of clustering was weaker.By comprehensive consideration of analysis results of the two kinds of marks,the strains belonging to two groups could be distinguished clearly.To sum up,it is feasible to apply the molecular markers to distinguish different strains of Lentinula edodes and Pleurotus,and their genetic similarity coefficient could be reduced accordingly along with increasing of the genetic distance among strains.
