| In this paper,the article deals with the pollution and some key factors which affect the anther culture of castor,such as basic medium,the hormone and the concentration of hormone,the sucrose and concentrate of sucrose,the concentrate of agar,the concentrate of active carbon,the time of low temperature treatment,high temperature treatment,the density of inoculation and so on.The result show:Buds were sterilized 75%ethanol(30s),followed by 0.1%HgCI2(3min),and fished fifth times with sterile water.The frequency of anther callus induction could be obviously improved in the condition of anther density over certain extent and the optimum density in the bottle of 50ml(about 20ml medium)was 3/5 bud per bottle.Although anther callus could be obtained from anther inoculated on MS,NB,N6 and B5 mediums,the MS medium was more suitable than other mediums.The buds was treated with low temperature forld,3d,5d,7d,9d respectively, the result shows that the percemtage of embtyoid formation increases obviously.The inoculated ather was treated with high temperature for 24h,48h,72h respectively,the result shows,after the appropriate high temperature and the time of high temperature treatment,the percentage of the embryoid formation enhances remarkably.The anther callus induction frequency between medium supplemented with sucrose and medium supplemented with maltose had not apparent difference,that indicated that sucrose and maltose had the same function.It is found 3%sucrose is the best concentrate for anther culture.The optimum bud induction medium the PGR with 1.5mg/L 6-BA and 0.5 mg/L NAA.It is found 7g/L agar is the best concentrate for anther culture.It is found 750mg/L proline is the best concentrate for anther culture.It is found 750mg/L Inorganic salt is the best concentrate for anther culture.100mg/L agar of medium was the most suitable agar content culture of alfalfa.Addition of certain activated charcoal(AC)were contributed much to tissue culture because AC could absorb harmful materials.
