RAPD, SRAP, SCAR were used to detect a total of 109 field Lentinual edodes strains collected from Wuping Longyan in Fujian province, Yunnan province and Sichuang province. Genetic polymorphism and relationship were studied on electrophoresis map with the cluster analysis of DPS,NTSYS. The results show that the average differential coefficient is 0.4348 among the strains tested, ranging from 0.0252-0.9630. Some tested strains are differed significantly. Geography Pricipal Coordinates Analysis showed that the strains tested were divided into 2 parts according to eastern and western regions of China. It shows that resource is regional distribution as a result of longitude variations of China. Main cultivars are basically outside the group of field strains. The results of the Principal Components of agronomic characters of tesed strains were as follow: the first Principal Components,which might be regarded as yield and fine fruit body morphology component factor(72.93%),growth rate proportion factor(15.02%).The two account for 86.07% among the varieties.At the same time, L0212 with high yield, fine fruitbody morphology,good turning color and L0249 with quick mycelial growth were chosen to make hybridization in accordance of the two Principal Components and distant relationship. Another producting mushroom was taken in order to select the ideal strain with hybridization strains which had clamp connection during the process of detecting mating factor.Finally,L0249XL0212Y was screened with quick mycelial growth, high yield, big size of body fruit, thick pileus center thickness,good turning color which cerified by clamp connection ,antagonistic test and molecular experiment. The yield of L0249XL0212Y showed 32% heterosis over the mian strain L0071(939).
