For 117 strains of wild Lentinus edodes collected fromLongyan,Sichuan,Yunnan, we studied the classification and their genetic relationship according to antagonism. Besides,we also studid the mycelial growth rate and the activity of extracellular enzyme, mycelial growth rate and the activity of protective enzyme in different environmental stresses, the characteristics of fructification growth period and nutritional conditions four aspcets , to explore some special germplasm and to construct core germplasm. The main research content and results are as follows:1. The classification and genetic relationship of Lentinus edodes by somatic incompatibilityIn this experiment,117 strains were divided into 98 categories according to whether the antagonistic reaction happened or not. At the same time, The phenomenon of somatic incompatibility was divided into four categories which were fusion, weak antagonism, apparent antagonism with brown line and apparent antagonism without brown line. Converted these into"0,1,2,3"datas,then we made a cluster analysis for all strains and devided them into 15 relationship groups.2. Comparison of mycelial growth rate and the activity of extracellular enzymeComparing all wild germplasm resources to the main cultivation strain L0018,we found strains L0222,L0226,L0275,L0295 relatively grew faster ,and strains L0222,L0223,L0229,L0236,L0299,L0302,L0304,L0305 performed prominent. Besides,we also studied the relationships between mycelial growth rate and the activity of each extracellular enzyme.The results showed that there was a significant negative correlation between the activity of laccase and mycelial growth rate, a significant positive correlation among the activities of carboxymethyl cellulase, xylanase and protease,a significant positive correlation between the activity of laccase and that of xylanase.3. Comparison of mycelial growth rate,the content of malondialdehyde and the activity of protective enzyme in different environmental stresses Comparing all wild germplasm resource to the main cultivation strain L0018,we screened some strains with adverse-resistant characteristics. The prominent low temperature stress-resistant strains were as follows: L0194,L0238,L0241,L0298; The prominent high temperature stress-resistant strains were as follows: L0194,L0238,L0241,L0298;The alkali stress-resistant strains were as follows: L0199,L0295,L0234,L0242,L0247,L0293,L0297,L0298,L0301;The drought stress-resistant strains were as follows:L0275,L0195,L0200,L202,L0206,L0214,L0221,L0224,L0225,L0226,L0227,L0228,L0244,L0245,L0250,L0262,L0267,L0271,L0276;The prominent humidity stress-resistant strain was L0288.4. Comparison of the characteristics in fructification growth periodBy comprehensivly comparing the fruting time,the morphology of fruiting body, the fresh weight of single flower and the yield of all wild germplasm resource,we found that L0248,L0195 were best and L0237,L0238,L0239,L0255,L0262,L0274 were relatively good.5. Comparison of the characteristics of nutritional conditionsWe comparied four situations of the wild Lentinus edodes germplasm resource which were separately cultivated in the sawdust of Lauraceae, Eucalyptus spp, Cunninghamia Lanceolata and Pinus,and taked Lauraceae for control. The results were as follows: Cunninghamia Lanceolata and Pinus without pre-treatment were not suitable for cultivation of Lentinus edodes but Eucalyptus spp is suitable.Among so many strains,we found that it was better for L0195 to cultivate in the sawdust of Eucalyptus spp than in that of Lauraceae.
