Prunus mongolica Maxim. is a kind of landscape plant in the desert area and the desert steppe. It is important in soil and water conservation. And, it also be a kind of important soil retaining shrub dry area full of stone with a sunny exposure in Inner Mongolia. Now, It listed as National Gradeâ…¢protected plants, have become an endangered species. The Prunus mongolica Maxim. tissue culture is conducive to better using or studing that species. It also be providing an important technique support for protecting the germplasm and rapid propagating on Prunus mongolica Maxim.By selected explants, culture medium, plant hormones. The investigation indicates: The best explants is stems of one-month-old seedlings; The best culture medium is solid medium. And, The callus formation rate will be 87 percent if the concentrations of 6-BA and NAA both are 0.7mg?L-1. We got the same results in the repeat experiments; it shows that could be repeated very well. So, in other tissue culture experiments on Stems of young Prunus mongolica Maxim. plant, it will be good with the concentrations of 6-BA and NAA both are 0.7mg?L-1But the callus browning is a too serious to continuation of the experiment. The browning phenomenon of callus during the Prunus mongolica Maxim. tissue culture procedure has been researching by fifteen samples cultured the different time and the different conditions. The total phenols content, polyphenol oxidase (PPO) activity, superoxide dismutase (SOD) peroxidase (POD) activity have been measured by spectrophotometry. The results revealed that the higher SOD activity and the lower POD activity are the important factors for callus browning, besides of the total phenol content in general.
