In order to further improve the endangered plant Prunus mongolica Maxim,the tissue culture technology system,this article uses Prunus mongolica Maxim seed as the test material to study the effects of different parts of the seed on the induction of adventitious buds,and screen out suitable for the initial cultivation,adventitious multiplication,rooting culture The medium,hormone type and concentration,and the results show that:(1)Seeds treatment:After the seeds are hulled,place them in a clean bench after soaking for 12 hours,disinfect with 0.1%liters of mercury(HgC12)for 10 min+2%sodium hypochlorite(NaClO)for 6 min,and rinse with sterile water 4 to 5 times each After washing and peeling,Use the inoculation knife to cut 6 parts of the seeds needed for the experiment and connect them to the medium.(2)Seed primary culture:The mature embryos are cut into 6 different parts and inoculated into 5 basic mediums and 5 different hormone concentrations.The selected part VI was most suitable for inducing adventitious buds from mature almonds of Mongolian almond.MS was selected as the best basic medium.The most suitable hormone concentration combination was 6-BA2.5mg·L-1+NAA 0.4 mg·L-1.(3)Subculture:The robust seedlings cultured from the first generation were cut into stem sections,and the orthogonal test method of three factors and three levels was used to screen the optimal proliferation hormone concentration of 6-BA1.0 mg·L-1+GA30.1mg·L-1+IAA0.2 mg.L-1.2?3 strains and all transplants were used to select the weak seedlings in the primary culture,and 2-3 strains were selected as the best way to proliferate.(4)Rooting culture:The hormone IBA0.4mg · L-1 was screened for the best rooting effect,and the main root was thick with fibrous roots.The optimal basic medium was selected for 1/2MS;the optimal sucrose concentration was 30g·L-1;the optimal dark culture day was 5d.
