Lonicera macranthoides Hand.-Mazz.is kind of liane growing for many years.It belongs to Caprifoliaceae macranthoide;it's the same with other Lonicera with the beginning of the dry buds collectively.At a broader application of clinical medicine,is an important one of the traditional Chinese herbal medicines. Contains volatile oil,flavonoids,tfiterpenoid saponin,organic acids,and inorganic elements and so on,have antibacterial,anti-inflammatory,antipyretic,choleretic, hepatoprotective,hypolipidemic,hemostasis and other medical value.This paper presented the laboratory test method for rapid in vitro propagation and analysised antifungal activity of the callus alcohol extract from Lonicera macranthoides Hand.-Mazz.and compare antifungal activity with the extract from Lonicera macranthoides Hand.-Mazz plant cultivation.The stemss and leaves were used as explants,adding different concentration of plant hormone to carry out callus inducing,multiplication and buds differentiation,multiplication culture and roots production.In the callus inducation and multiplication culture,(1)The leaves of Lonicera macranthoides Hand.-Mazz of 4-5month were used as explants,(2)After the explant culture in dark a week,it culture in light at 28℃.In this way,the callus browing rate was reduced and callus inducation rate was higher,(3) the optimal medium for callus induction was MS+2,4-D1.0 mg/L+6- BA0.5 mg/L+ NAA0.3 mg/L,(4) the best medium for callus proliferation was MS+6 - BA1.5 mg/L +NAA0.5 mg/L,the 7-9 generation of callus was best.2 In the buds inducing,multiplication culture,(1)the best medium for buds differentiation was MS +6 - BA1.0 mg,/L+NAA0.1 mg/L+KT0.2 mg/L,(2) the optimal medium for buds multiplication was MS +6 - BA1.0mg/L+NAA0.1 mg/L,and the optimal sucrose concentration was 30g/L.(3) and the best rooting medium was 1/2MS+IBA3.0 mg/L+carbon200 mg/L.The antifungal effects of the alcohol extract from Lonicera macranthoides Hand.-Mazz callus to Pestalotia funerea,Rhizoctonia solani Kuha.,Curvularia lunata(walk) Boed,Pyricularia oryzae Cav.and Gibberelle zeae(Schw.) Petch.5 kinds of plant pathogenic fungi were studied by using inhibiting fungi tests and electron microscope technique.The results showed that:The alcohol extract from Lonicera macranthoides Hand.-Mazz callus could obviously inhibit hyphal growth at 20 mg/mL.to 5 kinds of fungi the values of IC50 indicated that the alcohol extract from Lonicera macran- thoides Hand.-Mazz callus had highest inhibitive effect on Pestalotia funereaand and Rhizoctonia solani Kuha.and lowest inhibitive effect on Gibberelle zeae(Schw.) Petch.in 5 kinds of fungi.The hyphal cell wall disappeared and protoplast was dissolved by observation on electron microscope.These results suggested that the Lonicera macranthoides Hand.-Mazz callus had stronger antagonistic activity on 5 kinds of fungi,than the extract from Lonicera macranthoides Hand.-Mazz plant cultivation.
