The plant of Lycoris radiate.Herb is a novel starchy feedstock that contains various alkaloids and polysaccharide. The demand for alkaloid, which have functions for human's central nervous system and cardiovascular system, in field of clinical medicine and new medicine exploration is tremendous. Since the polysaccharide contained in the plant has solid potentials in guaranteeing human health due to its unique activity and nature originality, the research of which tends to be quite valuable for its medical application. Additionally, the fresh bulb of this plant contains nearly 20% starch that potentials can be used for ethanol production. This paper discussed the practicability of bio-refinery the plant of Lycoris radiate.Herb comprehensively. The researches on extraction of alkaloid, polysaccharide and the ethanol fermentation post-extract process were conducted, respectively, on the study basis of converting the starch component into ethanol with high efficiency, all aiming to obtain the optimum combined yields of alkaloid, ethanol and polysaccharide.This study starts from the research of the fluid characteristic of slurry from bulb (SFB) of Lycoris radiate.Herb, and its potential as being applied in ethanol fermentation was reviewed. It's demonstrated that SFB is typical viscoelastic fluid with dominant viscous. Effective viscosity reduction was achieved after the slurry mash being subjected to enzyme ofβ-glucanase, xylanase and pectinase, respectively. The simultaneous saccharification and fermentation adopting the SFB pretreated by pectinase as substrate was conducted, which gives rise to an ethanol yield of 1.9% (w/w) as the highest at the end of 70h by Saccharomyces cerevisiae. It's assumed that some metabolic pathways of yeast may be inhibited by the alkaloids contained in the bulb mash.The scheme of extracting the alkaloid within the lab and the ethanol fermentation using post-extraction solid was studied and the combined yield of alkaloid and ethanol in this process was assessed. The scheme was determined: the sample was reflux condensed using 95% ethanol at constant temperature of 85℃, air pump filtration followed and the liquid phase was collected, condensation, dissolution by methanol, finally the colorimetric determination at 289nm with methanol as control was measured. The batch ethanol fermentations using the dried residue obtained after the alkaloid saturating for 1,2,4,6,8h from the powder of Lycoris radiate.Herb, respectively, which were yielded through drying, grinding the fresh bulb. The results shown that the highest combined yield of 1420mg total alkaloids and an ethanol yield of 3.1%(w/w) was achieved by the test group of 4h.The ethanol fermentation adopting the waste residue from Lycoris radiate.Herb provided by a factory was studied. This part of work can be considered as the fermentation run after the alkaloids undergo the very different extraction scheme invented by the factory. It's demonstrated that the waste residue contains 41% starch and an ethanol yield of 5.8% (w/w) at the end of 48h, accounting 85.5% of the theoretical was achieved by the screened wild yeast, which performs more superior than the industrial yeast both in terms of the swiftly sugar consumption and high productivity. The extraction scheme of alkaloid invented by the factory tends to be more appropriate for the latter ethanol fermentation run. In addition, the extraction work of polysaccharide contained in the waste residue and ethanol fermentation using the post-extraction mass was committed. The extraction scheme using hot water was determined: the sample under substrate to water ratio 1:10 was maintained in water bath constantly at 90℃for 1.5h prior to centrifuged, the soluble starch in the supernatant obtained was hydrolyzed followed the addition of 95% ethanol with a volume of two times more than the supernatant, be kept overnight at 4℃, then the precipitations were collected by centrifuge and were dried to constant weight at 80℃under atmospheric pressure. An optimum combined yield of 9.5% crude polysaccharide and ethanol yield of 2.4%(w/w) was achieved after waste residue being extracted for one time by hot water, which decrease with the extraction times.The comprehensively bio-refinery of alkaloid, polysaccharide, starch consists in the plant of Lycoris radiate.Herb was preliminarily realized in this work, which is easy to maneuver and the potential for this process is numerous.
