| The objective of this study was mainly to evaluate the removal efficacy of mycotoxins (AflatoxinB1(AFB1),Zearalenone(ZEN),Deoxynivalenol(DON)) by sodium humate in vitro.This study included two experiments.The first experiment was mainly to study the detoxification of pure mycotoxins by sodium humate;at the same time,the influence of ELISA,interaction time,the amount of sodium humate,solution PH,initial concentration of mycotoxins(AFB1,ZEN,DON) and with or without feed(AFB1<1ng/mg, ZEN<10ng/mg,DON<10ng/mg) on the detoxification of mycotoxins(AFB1,ZEN,DON) by sodium humate were also studied.Second,we investigated the detoxification of mycotoxins in naturally contaminated corn by sodium humate and the stability of the sodium humate-mycotoxin complex in the presence of simulating intestinal solution.The results showed that:(1) Between the effect of detoxification by sodium humate and the interaction time existed in a significant time-response relationship(P<0.05).And the best interaction time was 60min.(2) Between the dosage of sodium humate and the effect of detoxification existed in a significant dose-response relationship(P<0.05).And the best detoxification dosage of sodium humate was 10mg(2.5mg/ml).(3) Solution PH was the main influence factor of detoxification.In this study,from PH 7 to PH 8,the detoxification of AFB1,ZEN and DON would reach maximum.The maximum rate of detoxification would reach up to 90.83%,50.08%and 16.98%, respectively.(4) There was a significantly linear correlation between the detoxification of mycotoxins by sodium humate and the concentration of mycotoxin(P<0.05).The detoxification of AFB1 and ZEN by sodium humate occurred in polylayers,the data was fitted better to Freundlich equation;while the detoxification of DON by sodium humate occurred in monolayer,the data was fitted better to Langmuir equation.(5) Sodium humate mixed with feed removed AFB1(ZEN,DON) with the same efficacy as in buffer alone(P>0.05),indicating that the coexistence of fresh feed have no significant influence on the removal ofmycotoxins.(6) The AFB1(ZEN,DON)-sodium humate complex was very stability in methanol, PBS and chloroform;the rates of desorption were all less than 20%;the rates of desorption were all less than 10%especially in chloroform.(7) In the solid state,the rates of detoxification of AFB1,ZEN and DON in naturally contaminated corm by sodium humate were 6.54%,41.32%and 29.07%;and in the simulating intestine solution,they were 52.32%,38.66%and 14.41%,respectively.(8) The AFB1(ZEN,DON)-sodium humate complex were very stable in the simulating small intestinal solution and the desorption rates were all less than 10%.These results suggested that sodium humate have a certain detoxification effect of common mycotoxins(AFB1,ZEN and DON),especially to AFB1 and ZEN,and the effect of detoxification were all better in alkalescent solution than in acidity.The sodium humate-mycotoxin complex was very stable in the simulating intestine solution and which were mainly due to non-physical adsorption. |
