Minature pigs had extensively been used as the experimental animals on frontiers of science researches, such as medical animal models, transgenic animals, clone, organ transplantation, embryonic transplantation and embryonic implantation. The satisfactory anesthetic course was needed for miniature pigs to complete these experiments. Specific combined anesthetic for miniature pigs-XFM was invented by balanced anesthesia theory and physiological characteristics of miniature pigs. Effects of XFM on blood pressure, heart rate, blood cytokines, neuropeptide, and neurotransmitters were researched in this study, in order to investigate the effects of XFM on hemodynamics and its mechanism in miniature pigs.Chinese experimental miniature pigs were injected combination anesthetic for miniature pigs-XFM, and dynamic monitoring of serum NO, plasma ET, PGI2, TXA2 , R-A-A-S, ANP, CGRP, NPY, NT, and to investigate the effects of XFM on hemodynamics and its mechanism in miniature pigs.Fourteen Chinese experimental miniature pigs were injected XFM 0.15mL kg-1, and blood was collected through precaval vein at 0, 5, 10, 30, 45, 60, 80, 100 and 120 min, at the same time SBP, DBP, MAP and HR were monitored. Blood samples were let in tubes which included different gelation inhibitors and enzyme inhibitors and carried out pretreatments, then measured by colorimetry and radioimmunoassay.The results showed that after injected XFM 0.15mL kg-1, miniature pigs demonstrated good anesthetic effect, quick induction of anesthesia, suitable maintain anesthesia time, calmer recovery; and better analgesia, sedation, muscle relaxation effect. Significant hemodynamic changes were found in miniature pigs during this experiment, the results showed that SBP, DAP, MAP and HR were demonstrated the trend of rising and then decreasing, and the changes were in range of physiological tolerance.After being injected XFM, multivariable analysis of the correlation between the hemodynamics changes and serum NO, plasma ET, 6-keto-PGF1α, TXB2.The results demonstrated that significant or extremely significant correlation between the hemodynamics changes and plasma ET, 6-keto-PGF1α, TXB2. And NO was not correlated with hemodynamics changes. The results showed that plasma ET, 6-keto-PGF1αand TXB2 participated in changing of hemodynamics which were caused by XFM in miniature pigs. After being injected XFM, plasma PRA, AⅡ, ALD and hemodynamics changes demonstrated consistent trend. And significant or extremely significant correlations were observed between R-A-A-S and hemodynamics changes. The results showed that R-A-A-S participated in changing of hemodynamics which were caused by XFM in miniature pigs.After being injected XFM, multivariable analysis of the correlation between the hemodynamics changes and plasma NPY,NT,CGRP and ANP. The results demonstrated that there was negative correlation between ANP and SBP, MAP, and significant negative correlation were observed between the ANP and DBP. And there was negative correlation between CGRP and SBP, DBP, MAP. NPY and NT were not correlated with hemodynamics. The results showed that the plasma ANP and CGRP participated in changing of hemodynamics which were caused by XFM in miniature pigs.This research showed that hemodynamic changed obviously after being injected XFM 0.15mL kg-1 and significant or extremely significant correlation were observed between hemodynamic changes and plasma ET, 6-keto-PGF1α, TXB2, R-A-A-S, CGRP and ANP. Therefore, the change of plasma ET, 6-keto-PGF1α, TXB2, R-A-A-S, CGRP and ANP were the main factors of changing of hemodynamics which were caused by XFM in miniature pigs.
