It is,Matricaria chamomilla L.,a kind of annual or perennial herb, is with great exploitative value for its volatile oil. This study establishes regeneration system of Matricaria chamomilla L. through tissue culture , chooses the best extraction process of volatile oil through orthogonal test , uses GC-MS to analyse the chemical constituents and also makes some researches on bacteriostatic activity,antiinflammatory and evacuation of free radicals in order to offer theoretic basis and technological support for the plantation,exploitation as well as exploitation of Matricaria chamomilla L.1. The foundation of regeneration system of Matricaria chamomilla L. The experiment studied effects of different antiseptic ways on seed germination and chose the best way among them; with terminal buds of sterile sprouts,effects of different basic culture medium,phytohormones as well as their combinations and sucrose concentration to the adventive buds'proliferation were studied and effects of different phytohormones as well as their combinations to the adventive roots were studied too with the purpose of choosing the most suitable culture medium and building an effective and quick isolated regeneration system.The outcomes were as follows: Using 75% alcohol for 30seconds,then 3.5% sodium hypochlorite for 15 min,finally 2% sodium hypochlorite to the sterilization of seeds as the best.the best midium for the proliferation of adventive buds: MS+6-BA1.5 mg·L-1+NAA0.5 mg·L-1;the concentration carbon source,3% of sucrose showed the best;the best midium for rooting: MS+IBA1.0 mg·L-1+NAA0.5 mg·L-1+6-BA 0.2 mg·L-1;the best groundmass for transplantation: vermiculite: perlite: charcoal (1∶1∶1)。2. The extraction of volatile oil: steam distillation method was used and orthogonal test including four main factors such as the time of extraction,the quantity of water input,the size of material and the time for soaking was taken to choose the best way of extraction.The result showed that soaking for 6h,water of 8 times,10h extraction for 10h.and the middle size were the best conditions for extraction .3. Using GC-MS to study the chemical constituents:the result showed: about 14 compounds were tested and among them the concentration of 12 compounds belonging to olefine,enol,acid and aldehyde reacheed 96.3%; the concentration of alkane were 1.48%;acid 1.7%; aldehyde 1.54%. The three highest constituents were 2H-Pyran-3-ol.terahydro-2,2,6-trimethyl-6-(4methyl-3-cyclohexen-1-yl)-,[3S-[3. alpha,6.alpha.(R@)]]- reaches 31.65%,2-Furanmethanol,tetrahydro-.alpha.,.alpha.,5-trimethyl-5- (4-methyl-3-cyclohexen-1-yl)-,[2S-[2.alpha.,5.beta.(R@]]-reaches 18.66%,2-(4-methylenecyclohexyl)-1-propanol reaches 18.18%。4. The antopxidation of volatile oil: volatile oil had better reducing power. When the concentration was 0.2mL/mL, the reducing power got 50.3%; when the concentration of volatile reaches 0.4mL/mL,the inhibition ratio for the lipid peroxidate was 47.9%;when 0.6mL/mL,the clearance of DPPH was 51.1%, hydroxy radical 54.4%. The concentration and antopxidation of volatile oil acted as dose-effect relationship,the higher the better.5. Bacteriostasic activity in vitro: Cup method was used to check the inhibitory effects on general fungi and bacteria. The result was that:the effect on bacteria was better and effects on Bacillus coli and Bacillus subtilis are better ,compared to Staphylococcus aureus,;MIC on Staphylococcus aureus was 50%, Bacillus coli and Bacillus subtilis both were 25%. The effect on head mold were apparently higher than blue mould and Aspergillus niger. MiC on head mold was 6.25%,blue mould 50%, Aspergillus niger 75%.6. anti-inflammatory effects in vivo:The study of the inhibition to foot suellingof rats caused by egg white ,the proliferation of the granulation caused by tampon transplantation and the ear suelling caused by xylene and the influence on the con-centration of TNF-αand IL-Iβfrom inflammatory exudate were included in this st-ep.The results showed different dose of volatile oil shows different inhibition towards foot suelling. Dose group in 0.5h had extremely significant difference with the model group(P<0.01),1h suelling had significant difference with the model group (p<0.05),large dose group in 0.5h,1h,3h had significant difference with the model gr-oup(p<0.05).Volatile oil had certain effects on acute inflammation caused by egg w-hite.All the dose group had certain effects on the proliferation of the granulation,The pure gravity of this group has significant difference with the model group,among them the wet gravity of granulation in large dose group had significant difference with the model. Volatile oil showed certain inhibition on the proliferation of the gr-anulation of chronic inflammation.All dose group had different inhibition to the ear suelling,large dose group had extremely significant difference with the model group(P<0.01).The study shows volatile oil had certain effects on suelling and exsudationcaused by acute inflammation.
