The dual-purpose peaches and the nectarines which were the selections of Institute of Agricultural Integrated Development, Beijing Academy of Agriculture and Forestry Sciences were studies by genomic DNA-AFLP technology. By construction of Fingerprinting Map of peach varieties and studies on crisp/melting character of peach fruit by AFLP Markers, the theatrical and technological foundations were established for variety identification and intellectual property protection of peaches, as well as auxiliary selective breeding by molecular mark and so on. The findings are as follows:1) 23 dual-purpose peach varieties such as Prunus persica var. nectarina cv. Jinchun were studied by genomic DNA-AFLP technology.4 primer pairs (E-AAT/M-ACT,E-AAT/M-CTG,E-ACA/M-CTGå’ŒE-ACA/M-CTT)with rich polymorphism and high quality were selected from 64 ones.127 DNA fragment bands were amplified; including 64 polymorphic bands and polymorphism percentage was 50.4%. The amplified polymorphism bands and polymorphism proportion were relatively low because the varieties have closer genetic relationship. The fingerprint map of 23 dual-purpose peaches was constructed using the selected 4 primer pairs, so it will lay certain foundation for varieties identification and protection of dual-purpose peaches.2) 14 nectarina varieties such as nectarina cv. Xinchun were studied by genomic DNA-AFLP technology.4 primer pairs (E-AAT/M-ACT,E-AAT/M-CTG,E-ACA/M-ACTå’ŒE-ACA/M-CTT)with rich polymorphism and high quality were selected from 64 ones.76 DNA fragment bands were amplified; including 60 polymorphic bands and polymorphism percentage was 78.9%. The amplified polymorphism bands and polymorphism proportion were relatively high because the varieties have differant genetic relationships. The fingerprint map of 14 nectarina varieties was constructed using the selected 4 primer pairs, so it will lay certain foundation for varieties identification and protection of nectarina varieties.3) In this research molecular markers to crisp/melting trait in peach rriuit with AFLP technique coupled with BSA method in 15 population hybrid from the selfs of nectarina cv. Qing 27-59. Among 64 pairs of AFLP primer combination,23 pairs produced polymorphism between three bulks and their parent. The result of individual amplification showed that only three of them amplified specific fragments to crisp/melting trait. The specific fragments derived from primers combination E-ACA/M-CTT and E-AAT/M-ACT.Thie study would be important for further SCAR marker conversion.
