Pueraria thomsonii Radix is a leguminous plant Pueraria thomsonii (Benth.)’s dried root. It has sweet, acrid and cool, which could be used in the treatment of relieving muscles to expel heat, encouraging production of body fluids to extinguish thirst, lifting yang to relieve diarrhea, clearing and activating the channels and collaterals. It is a traditional medicinal and edible plant. In recent years, its medicinal and edible value has been paid more and more attention, especially in the development of the health products. Therefore, it has broad development value and broad market prospects.This subject used the buds and shoot tips as explants, of which Qinba2from Dazhu were inoculated in starting medium after disinfected. The results indicated that the best starting medium was MS+6-BA2.0mg/L+IAA0.2mg/L; The best multiplication medium of cluster shoots was MS+6-BA0.5mg/L+KT0.1mg/L+IAA0.05mg/L; The results showed that taking the stem as explants to induce the callus was better than leaves and leafstalks, and the best method of callus culture was that explants were cultured in the darkness until the callus began to grow, then they were cultured in the light (illumination is12h/day, light intensity is12001ux, cultural temperature is25±℃), the best callus induction medium was MS+6-BA+IAA0.2mg/L; The best rooting medium was1/2MS+IAA1.0mg/L; Selected the robust growth of tube seedling transplanted to nursery bed after seedling adaptation (nutrition soil: vermiculite:pearl salt=3:1:1), the survival rate could be obtained more than95%. This method can be quickly established the propagation system of Pueraria thomsonii (Benth.).This subject studied in artificial seeds, which were made by the buds oi Pueraria thomsonill (Benth.)’tube seedling. The results showed that the best manufacturing conditions of artificial seeds was the reaction time8-10min of sodium alginate3%in calcium chloride2%; Artificial endosperms was MS1/2+IAA0.5mg/L+sucrose30g/L of the artificial seeds were100%; The artificial seeds cultured in the medium better than culture dish. The contents of copper in Prueraria thomsonii (Benth.) were determined by flame atomic absorption spectrometry (FAAS), and the contents of lead and cadmium in Pueraria thomsonii (Benth.) were determined graphite furnace atomic absorption spectrometry (GFAAS), Under the optimum conditions, the detection limit of copper, lead cadmium were:0.03mg/L,0.243μg/L and0.012μ.g/L, the recovery is from97.26%to105.60%, and the RSD less than5%. The content of copper, lead and cadmium from different areas were in accordance with the national standard of Green Trade Standards of Importing&Exporting Medicinal Plants&Preparation. With0.2%magnesium nitrate and ammonium dihydrogen phosphate1%as matrix modifier, the ash temperature of cadmium and lead can be increased to500℃and800℃, and can improve the atomization temperature and reduce the matrix interference, This method is simple, rapid and sensitive.
