China is one of the origin country of lily resources, but breeding work of lily begins later, lily varieties which breeded by China and self-owned intellectual property rights are few, In order to speed up the breeding work in China, genetic relatives was analyzed by phenotypes and SSR genotype, and parent selection was studied with tens of introduced excellent cultivars as materials in this research..The results showed:1. Pollen of'Justina Zantrijus','Sheila','Elite'and'Lilium regale'storaged inside of desiccator at 4℃was the suitable condition, which ability of germination could keep 84,35,28 and 84 days separately; pollen of'Brunello'stored outside of desiccator at 4℃was the best way, which ability of germination could keep 70 days; however, the best storage condition of Lilium davidii var. unicolor pollen was stored outside of desiccator at-18℃, which vitality could keep 49 days. With the storage time increasing, the germination rate of lily pollen gradually decreased, but there were distinctly rising in some periods of storage process.2. Hormone of different concentration and proportion had different effects on the same kind of pollen,hormone of same concentration and proportion had different effects on the different kind of pollen.3. Germination rate of pollens of Lilium davidii var. unicolor decreased gradually as increase of treated temperature, at the same treated temperature, the longer treatment time, the lower the germination rate of pollen. Germination rate of pollens of'White Fox'treated 1 hour and 2 hours at 35℃was lower than that treated the same time at 40℃,45℃and 50℃, when the pollens were treated 3 hours, the germination rate of pollens was the highest at 50℃. The pollens of'Sorbonne'couldn't stand up to high temperatures, their germination rate decreased quickly as the temperature increased.4. Cross-combination between Oriental lily hybrids could get fruit and embryo, but different cross-combination whose cross-compatibility were different, some of them could not get embryo, The cross-compatibility of'Willeke Alberti'was the best as female materials; the cross-compatibility of'Topwhite Zanler-white'was the poorest as female materials. Hybrid embryo by distant cross-combination was not obtained.5. The embryo of different cross-combination needed different concentration and kinds of hormone, the optimal culture medium was MS+0.1mg/L NAA+0.5mg/L6-BA+40g/L sucrose+6g/L agar for hybrid embryo of'Sheila'×'Francia'and MS+0.01 mg/LNAA+1.0 mg/LBA+40 g/L sucrose+6 g/L agar for hybrid embryo of'Ceb Dazzle'×'Elite'.6. The research of genetic relatives based on phenotypes and SSR genotype indicated that the matrixes of them were inapperant minus correlation, the coefficient was-0.0022(p=0.9737). It proved that the character of phenotype we chose could not correctly reflect the genetic variability of materials with a near relationship.7. From the results of cross-combination, we could see that there was no clear relation between cross-compatibility and the genetic distance based on SSR. But when the distance exceeded more than 1.0, no cross-combination could get embryo.
