Radix Et Rhizoma Salviae Miltiorrhizae is the dry roots and rhizoma of Salvia miltiorrhiza Bge. in Chinese traditional medicine. In order to utilize this plant resource more deeply, we have to study on the effective constituents of the fibrous roots of Salvia miltiorrhiza(Shangluo, ShaanXi). Firstly, the diffferent extracts of the ethanol extract from Salvia Miltiorrhiza Bge. were used to determine total phenols with Gallic acid as standard compound. Then the free radical scavenging activities of different fractions were measured with DPPH·method. Secondly, using 3 kinds of plant pathogenic fungi as testing fungi to examine the antibacterial activities of the alcohol extract of Salvia fibrous roots in vitro. Lastly, based on the known results, utilizing the photochemistry theory and modern isolation and purification approach and spectrum technology to purifying and analysing the active components in ethyl acetate and ethylether of alcohol extract of Salvia fibrous roots. The main results are as follows:1. The antioxidant activity of the ethanol extract from Salvia fibrous roots.The research indicated that, the phenolic contents in 5 different polar solvents of alcohol extract of Salvia fibrous roots have a significant differences, which the ethyl acetate has the highest content about 51.48%, and the n-butanol is 31.8%, the chloroform is 11.11%, the ethylether is 2.74%, the water-soluble substances is 1.69%. The antioxidative examine result shows that, the IC50 for scavenging free radical of every ectraction by DPPH·of the 5 parts have a significant differences, which the ethylether is 210μg/mL, the chloroform is 122μg/mL, the ethyl acetate is 6μg/mL, the n-butanol is 42μg/mL, the water-soluble is 10277μg/mL. The related examine results shows that the antioxidative effect of alcohol extract in every solvent of Salvia fibrous roots is determined by the phenolic compounds of them.therefore, ethyl acetate extract part of antioxidant activity is best, n-butanol take second place, and both of them were further researched.2. Antibacterial activities of the alcohol extract of Salvia fibrous rootsExperimental results show that, the antibacterial efficiency to 3 kinds of Plant pathogenic fungi have a significant differences when the examine consistency is 1mg/mL, which the ethylether had the highest antibacterial efficiency to 3 kinds of plant pathogenic fungi, and the anti-bacterial efficiency is 81.44%, 85.58%, 82.73% for Botrytis Cinerea Pers., Rhizoctonia soluni. and Glomerella cingulata.. So the ethylether is the best solvent for extracting the active compounds from Salvia fibrous roots.Moreover, the different consistency of ethylether have the different effect on antibacterial. The figures of EC50 in 72h are 0.3615, 0.4221, 0.2708mg/mL for Botrytis Cinerea Pers., Rhizoctonia soluni. and Glomerella cingulata., all of them express significant activities of ethylether which is worth doing farther research.3. Isolation and purification and structure analysis of the effective components in ethanol Salvia Miltiorrhiza Beg. fibrous roots.The constituents of the ethanol extracts from Salvia Miltiorrhiza Beg. fibrous roots were isolated and purified through thin layer chromatography, repeated column chromatography on silica gel and recrystallization, and 8 compounds were gained, which were:â… (12.2mg),â…¡( 3.1mg),â…¢( 42.5mg),â…£( 16.3mg),â…¤( 3.5mg),â…¥( 15.6mg),â…¦(8.7mg),â…§(6.7mg). it was identified by 1H-NMR, 13C-NMR and MS that compoundâ…¢was 9″,9?-dimethyl lithospe- rmate B, compoundâ…£was 9′,9″-dimethyl lithospermate, compoundâ…¤was 2-N-(2′-hydroxy- palmitoyl)-1-O-β-D-glucopyranosyl-4-sphingaenine, which was firstly isolated from S. Compoundâ…¦was Cryptotansh- inone.Compoundâ…§was Dihydrotanshinone I .This thesis found that the total phenols content in ethyl acetate extract of ethanol extract from Salvia Miltiorrhiza Bge. fibrous roots is the highest, and it is in the strongest antioxidant capacity, And antibacterial activity of petroleum ether parts is best. 8 compounds were gained from the ethanol extracts from Salvia Miltiorrhiza Beg. fibrous roots, and 2-N-(2′-hydroxy- palmitoyl)-1-O-β-D-glucopyranosyl-4-sphingaenine was firstly isolated from S. Miltiorrhiza. The research enriches the research on salvia miltiorrhiza Bge.and provides the theoretical for the further research on salvia miltiorrhiza.
