Salvia miltiorrhiza Bunge is a traditional Chinese herb plant, its roots are used medicinally.In this study, we investigated Salvia genetic diversity by genome SSR.As material,99-2,99-3,shanhuang and Shandong cultivated Salvia are used for the field trials design.which is about different varieties, planting density, growth regulators, fertilizer type, years of growth and harvest. We also studied the effect of cropping. Determined the content of the six ingredients (tanshinone I, cryptotanshinone, tanshinone IIA, protocatechuic acid, rosmarinic acid, salvianolic acid B) and the level of antioxidant activity of Salvia samples,since then search the optimum cultivation conditions of Salvia. The results are as follows:1. SSR polymorphism locis of Salvia miltiorrhiza analysis results show that:the genetic diversity of salvia miltiorrhiza populations is rich and the proportion of polymorphic locis is98.98%;19Salvia populations can be divided into eight groups. The genetic diversity of Shandong Yiyuan Salvia populations is most abundant and its average percentage of polymorphic loci was57.14%; the genetic diversity of99-3Salvia population is lowest with the polymorphic loci proportion only0.68%; the genetic diversity of wild populations of Salvia (50.34%) is higher than cultivated Salvia populations (41.35%).2. The content of the six active ingredients and the antioxidant activity are different in different varieties and the99-3varieties is the best; there is no significent difference of the ingredients contant and the antioxidant activity to the three density; repreduction by the seed breeding of Salvia is better than the root; Zhuanggenling can make the production of the root breeding Salvia increase38.45%, Duoxiaozuo can make production of seedling breeding Salvia increase14.19%, both significantly reduced the antioxidant activity of Salvia,but they had no significent effcet on active ingredient conntents; the production output and antioxidant activity of biennial Salvia are better than one year, but the content of ingredients in annual and biennial Salvia is no significant difference; active ingredients content and antioxidant activity are significantly different in different variety Salvia that harvest in different time, the’ fat-soluble ingredients is the highest in April and the water-soluble ingredients is the highest in November of Salvia99-2; fat-soluble ingredients is the highest in August and both the water-soluble component content and the antioxidant activity are the highest in June of Salvia99-3.3. Continuous cropping reduced Salvia production by21.78%and antioxidant activity also significantly decreased. Using microbial fertilizer can increase yield by21.67%; fertilization can make the antioxidation activity significantly improve but has no impact on the active ingredient contents of cropping Salvia.
