Study On Hybridization Pollination Of Lily

14 species of lily varieties belonging to Asiatic hybrids, Oriental hybrids and wild varieties, respectively, were selected for hybridization. Investigation on characteristics, examination of pollen vitality and collecting time, pollen storage, SEM observation on the pollen morphology, hybrid pollination and culture of the filial generation were conducted. The results were as follows:(1) The wild varieties had reversed flower shape and strong cold resistance. But their flowers were without fragrance; Asiatic hybrids had flower type of bowl and up-open and strong cold resistance and their flowers were without frabrance; Oriental hybrids had flower type of star-bowl and up-open. Their flowers had frabrance, but they had poor cold resistance.(2) The optimal culture medium was made up with 100g/L sucrose,20~30mg/L H3BO3 and 20~40mg/L CaCl2. The optimal cultivation temperature was 25℃. In the bloom day, the optimum time for pollen collection of Asiatic hybrids and Oriental hybrids was about ten o'clock, while the optimum time for wild varieties was about sixteen o'clock.(3) When the pollen was separately stored at 4℃for about 75 days, at-20℃for about 100 days and at-80℃for about 365 days, the pollen vitality had almost losed.(4) The results of SEM observation on the pollen morphology showed that the lily had different sizes of pollen grain, with the shape of spheroid or long spheroid with germinal furrow. The surface of pollen was mainly single row pilum net veins. But L. leichtlinii var. maximowiczii had single-double pilum net veins. The pollen had different sizes of mesh and murus particles arrangement, but'Algarve' and'Siberia' had large mesh. L. lancifolium,'Prato','Pollyanna', 'Yelloween','Siberia' had loose arrangemengt, the others arranged densely.(5) The results of hybrid pollination showed that in the 32 hybridized combination,11 hybridized combination obtained capsules, and 4 hybridized combination obtained embryo hybrid seeds. The seeds had a greater difference on shape, size, transparence and size of embryo. Reciprocal cross hybrid had a certain effect on the rate of capsules setting. The capsules setting rate could be promoted through cut-style pollination and pollen culture medium solution painting stigma pollination for hybridized combination with a certain affinity to some extent.(6) The results of culture of the filial generation showed that the optimal cultivation temperature for hybrid seeds of A2×A3 was 15℃. The light was the prerequisite for germination of hybrid seeds with sterile culture. The seed germination rate could be enhanced through adding a certain concentration of NAA and halving or no adding agar.40~50 days after pollination was a best harvest time for young embryo culture. The young embryo grew better on MS media with 60g/L sucrose and 0.5~1.0mg/L CH. MS media supplended with 0.1mg/L 6-BA was favorable to the growth of phyllome. MS media supplended with 0.3~0.5mg/L 6-BA was suit propagation. The young embryos were cultured in dark condition for 2~4 weeks and then transforming in light culture. Thus it was helpful to young embryo and hybrid seedling growth in late stage.
