Seabuckthorn belongs to Elaeagnaceae Hippophae. Elaeagnaceae Hippophae (Hippophae)has pharmacological effects on expectorant, Lee Lung, Stomach, spleen, blood, stasis.Few works had been done on the extraction of SDF from Seabuckthorn.Thus the possibility of the extraction conditions of SDF from Seabuckthorn by water extraction methods and the enzyme extraction methods were,for the first time,explored in this dissertation.Also,the effects of some variables on the extraction by two methods were comparatively examined and the combined extraction methods were also studied.Additionally,the structure and physicochemical properties of SDF from Seabuckthorn were analyzed.The Seabuckthorn contains that 68.34% of DF,include 64.14% of insoluble fiber and 2.2% of soluble dietary fiber;7.2% of water;20.02% of proteins;4.61% of fat and 5.7% of ash.It has been shown from the study on the extraction of SDF from Seabuckthorn by water extraction method.The optimal extraction agent PH valure,solid-liquid ratio,reaction temperature and reaction time were 7,1:35,70℃and 30min,respectively,under which the extraction rate of SDF from Seabuckthorn was 4.53%,and the pure rate of SDF reached 85%.Viscozyme L enzyme treatment dried Seabuckthorn.The optimal extraction agent pH valure,solid-liquid ratio,reaction time and ethanol precipitation were 5,1:25,120 min and 4 hours,under which the extraction rate of rate of SDF from Seabuckthorn insoluble fiber was 8.59%, and the pure rate of SDF reached 92%.The particles of the SDF prepared by enzyme were smaller,and the particles were irregular in shape under the electron microscopy.The result of UV analysis showed that not has protein, polypeptide and nucleic acid.The result of IR analysis showed that the characteristics of SDF were typical infrared spectra of polysaccharide.There were the characteristic absorption pesks of functional groups of polysaccharide.The characteristics of SDF were studied.The SDF were analyzed to be water holding capacity 4.818g·g -1,swelling capacity 5.506 g·g -1,water binding capacity 2.28 g·g -1 and cation-exchange capacity 0.575 mmol·g-1.Adsorption unsaturated fatty acids 1.769 g·g -1. Seabuckthorn SDF has more physiological active,can adsorption chelation cholesterol,bile acid in the intestinal.The growth-promoting action of Seabuckthorn SDF on Bifidobacterium was studied by on vitro assay.The results show that Seabuckthorn SDF oligosaccharide is an effective bifidobacterium growth-promoting factor,and the strain of tested Bifidobacterium grew significantly better in Seabuckthorn SDF media with 0.5%-1.0% Seabuckthorn SDF as the carbohydrate resource. Bifidobacterium is an obligate anaerobe.With this kind of germ,vareties of carbohydrate chould be fermented to formacetic acid and lacitic acid.As a result,the pH value is decreased. Bifidobacterium is an acid-sensitive germ,its growth would be remarkably depressed.
