Background Asthma is one kind of chronic airway inflammation characterized by eosinophil recruitment and mast cell response. Leukotrienes can have effects on many parameters on airway function in asthma, including the development and regulation of inflammation. Anti-inflammatory therapy is considered to be the most effective treatment of asthma especially the inhaled corticosteroids. Leukotrienes antagonist is one of new active therapeutics of asthma in recent years. Material and methodAnimal: Male Sprague Dawley rats weighting about 130 g were distributed randomized into 5 group: budesonide treated group (BUD), montelukast treated group (M), 1/2 budesonide and 1/2 montelukast treated group (1/2 BUD+1/2 M), normal control group (NC), ovalbumin sensitized, challenged group (OVA). BUD Group, M group, 1/2 BUD+1/2 M and OVA group were sensitized with 1ml normal saline, 3mg ovalbumin mixed within lOOmg aluminum hydroxide gel, while NC group were injected only with1ml normal saline plus aluminum hydroxide gel. In M group, montelukast was orally given, once daily for 7 days from the 8th day after OVA sensitized. In BUD group, budesonide was inhaled twice daily for 4 days from the 11th day. All groups were challenged with ovalbumin in the 14* day and were killed in the 15th day, and blood, bronchoalveolar fluid (BALF) and bone marrow were collected for counting the number of white blood cell and eosinophil. Keep serum and the supernatant of BALF after centrifugation at -20 癈 ready for detect the level of IL-5 and IFN- y.Research design: Cell counts and cell differentials after Wright-Giemsa stain were made. Serum and BALF were collected to detect IL-5 and interferon gamma level. Then the lungs were cut to make slides.Statistics: Results are presented as the mean D. If the data were analyzed to be normally distributed variables, ONE WAY ANOVA were used in the same group, while those of different group using LSD. Manny-Whitney test in non-parametric test were used in data of differently distributed variables. PO.05 is considered to be statistically. ResultsWBC and eosinophil counts in BALF: WBC counts of BUD group (P<0.05), 1/2 BUD+1/2 M group (PO.01) and NC group (PO.01) were significantly higher than that of OVA group while M group had no difference vs. OVA group. Eosinophil counts of BUD group (PO.01), M group (P<0.05), 1/2 BUD+1/2 M group (PO.01) and NC group (PO.01) were all significantly higher than that of OVA group. There is significant difference between M group and NC group, but no difference were found in BUD group and 1/2 BUD+1/2 M group vs. NC group. It might be concluded that budesonide usealone or combined use of small doses of budesonide and montelukast can well inhabit the infiltration of WBC or eosinophil in the lung, but montelukast use alone have less efficacy.WBC counts in blood: There are no significant differences in WBC counts in blood among all the groups except the 1/2 BUD+1/2 M group vs. NC group. WBC and eosinophil counts in bone marrow: WBC counts in BUD group and NC group are significantly different from that of OVA group, and eosinophil counts in all groups have significantly difference compared with OVA group. It means that all applications can affect the differentiation and mature of eosinophil in bone marrow.Results of IL-5 and interferon gamma levels in serum and BALF: Serum IL-5 levels in 1/2 BUD+1/2 M group and NC group are significantly lower than that in OVA group. No significant difference is found between the BUD group and OVA group. Serum IFN-y in each group is significantly high vs. OVA group. There is no significant difference of serum IFN-y between the two treated groups. The levels of IL-5 in BALF of the two treated groups and NC group are significantly lower than that of the OVA group and the levels of IFN-y in BALF are one the contrary.Results on lung slides: There are significant eosinophil infiltration of the lung in OVA sensitized group, especially in the area among the bronchioles and small vessels and the phenomenon of bronchio...
