Objective (1 )Study on on superoxide anion free radical in vitro and try to find out the mechanism of ShengMai on free radical. (2)Use the cerebral ischemia and reperfusion model in gerbil to observe the anti- oxygenation of ShengMai and its influence on the apoptosis of neuron after ischemia. (3)By determining SOD and LPO ,observing the configuration of neurocyte and detecting the expression of p53 Bc1-2. NF- K B after the ischemia and reperfusion,the possible mechanism of the influnece of ShengMai on ischemic injury should be demonstrated. (4)Applying the ShengMai and ChuanXiong in the patients with acute cerebral infarction and demonstrating the therapeutical effect of ShengMai on the ischemic cerebrovascular disease. Methods (1) The superoxide anion radical which produced in alkaline dimethyl sulfoxide (DM50) system was directly measured by electron paramagnetic resonance (EPR). The signal of drug group and control group was calculated. (2)40 gerbils were randomized into five group of eight animals each for sham group, ischemialreperfusion group and ischemialreperfusion administration group in which ShengMai was administered at a dose of 2OmIIkg through abdominal cavity injection.The decollation was performed after 24 hours and the tissue of prosencephalon was obtained.The activity of SOD and content of LPO in the brain homogenate were assaied.The HE, TUNEL stainning and immunostainning with antibodies to p53, Bcl-2 and NF-kB to the slices of gerbil brain were performed after fixing and embedding these tissue. (3)70 patients of acute cerebral infarction were chosen within 72 hours and were randomized into three group which were control group ,ChuangXiong group andShengMai combining ChuangXiong group.The assessment to neurologic impairmentand curative effect were performed PretheraPy and post-tTedrient.(4)The value of K and R in each group of ShengMai were calculated in the part ofexperiment in vitro.While in the part of animal experiment,the activity of SOD, thecontent of LPO,the number of aPoptosis cell and the degree of immunohistochemistrydyeing of p53' Bcl-2. NF- K B were ana1yzed by the analysis ofvedance.In the clinicalpart xz and t test were aPpIied in the gemeral data,the analysis of variance was aPpliedbetWeen three groups and the Ridit in the curatic effect between theraPeu- tical grouP.Result8(l)Shengrnai increased suPeroxide anion radical when its concentration was less than40%,while decrease superoxide anion radical When its concentration was more than60%.(2)The activity of SOD decreasing,the content of LPO increasing,the positive cell ofMEL dyeing increasing and the varing degree of immunohistochemistry dyeing ofp53, Bcl-2 and NF-kB in the ischemia/reperfusion group had significanly statisticaldiffeence comparing with sham group(p<0.0l).While in the ischemia/reperfusiontheraPeutical grouP had not(p>0.05).There was a significanly negtive correlationbetWeen the activity of SOD,expression of Bcl-2 and the number of aPoptosis (P<0.0l),while a positive one between the content of LPO,the degree of immunohistochemistrydyeing of p53, NF- K B and the number of aPoptosis(p<0.0l).(3ap curative effect of ShengMai combining ChuanXiong had significanlyStatstical difference comParing with the ChuanXiong grouP(P<0.05).Conclu8ion(l)ShengMai could elirnnate suPeroxide anion radical When its concentration washigher,Wh1e promote when lower.What ShengMai has dual effect according itsconcelltration indicated the complexity of the traditional Chinese medicine whendifferent comPonent act in coordination..(2) ShengMai could protected the brain tissue throu...
