People suffer from the ionizing radiation of the high radioactive background substance in the industry, medical treatment and environments. Radiation could do harm to immunity and cause the declination of the body resistance to diseases and so that arouses the possibility of mutation and cancer. To a large degree, the radiation damage is mediated by the large amount of radical ionized by water. Radicals could injure all kinds of biological large molecules and membranes, causing the lost of cellular function, the turbulence of cell cycle and the instability of heredity. Subsequently, there's great concern on the application of traditional Chinese herbs having anti-oxidation and anti-radical effect in the prevention and cure of radiation damage. Saussurea involucrate Kar.Et Kir. which has been used for a long long period, has a wide pharmacological function and shows very good anti-oxidation and anti-radical effect. In this paper, we systemically analyzed the anti-radiation damage effect of the water extract of Saussurea involucrate Kar.Et Kir. and its possible function mechanism, by animal experiment, hydroxyl radical clearance test, reverse transcription polymerase chain reaction of the gene P21 which regulates the cycle period of the cell, and Giemasa staining dicentrics acentrics. The results showed that the water extract of Saussurea involucrate Kar.Et Kir. would generally prolong the existence time of the mice injured by radiation, increase the surviving rate, and significantly enhance the transform ability of the T lymphocyte in the mice spleen so as to improve the immunity function. It has an enormous clearance effect on hydroxyl radical generated by ionizing radiation which amounts to a degree of 60%. It could also inhibit the mutation rate of human peripheral lymphocytes chromosome aberration caused by ionizing radiation, and has little influence on the quick expression of P21 gene during radiation damage. All these results suggested that the water extracts of Saussurea involucrate Kar.Et Kit. could efficiently prevent radiation damage, and this effect may be achieved by clearing the radicals, esp. Hydroxyl radical which is the most reactive and of the greatest damage, produced in the radiation damage, preventing their attacks towards the large biological molecules such as DNA and proteins. It could also regulate the cell cycle by arrest G1 and relaying S after the radiation damage, so that the cell could have enough time to do normal repairing, or induce apoptosis of severely injured cells which could not be repaired to guarantee the heredity stability and reduce mutation caused by ionizing radiation and the incidence rate of cancer. This research offers theoretical basis for the application of Saussurea involucrate Kar. Et Kir. in the protection of radiation damage and effect of anti-radical and anti-oxidation.
