Objectives 1. To explore the advantage and disadvantage of the Artificial method and Automatic scan system of chromosome karyotype method for peripheral blood lymphocyte aberration testing. 2. To explore the long-term and low-dose ionizing radiation's effects to the aberration of the radiation workers' peripheral blood lymphocytes.Methods 1. 167 radiation workers' peripheral blood samples were analysed using Artificial method and Automatic scan system of chromosome karyotype method, and further statistical snslysis was carried out for the detection data. 2. 898 radiation workers'peripheral blood samples and 381 non-radiation workers' peripheral blood sampales were collected, and then lymphocyte aberration sample positive rates, MF and CAF of the radiation workers' peripheral blood lymphocyte was statistically analyzed according to the radiation workers with different types, genders and working age.Results 1 (1) 167 samples were detected both in artificial group and scanning group,and the difference of lymphocyte aberration sample positive rate between two groups were not statistically significant (P>0.05). (2)167000 lymphocytes were detected for MF both in two groups. The MF in Scanning group was 2.69‰ significantly higher than 1.95‰ in Artificial group (P<0.05). Then 33400 lymphocytes were detected for CAF between two groups. The CAF in Scanning group was 19.46(?) significantly higher than 5.39(?) in Artificial group (P<0.05). 2 (1) Analysis of the radiation workers with different types: The industrial group and medical group had a significantly higher lymphocyte aberration sample positive rate (7.49% and 8.42%), MF (1.84‰ and 1.70‰)and CAF (4.25(?) and 5.20(?)) than the corresponding control groups (lymphocyte aberration sample positive rate: 1.49% and 1.12%, MF: 0.37‰ and 0.36‰, CAF: 1.73(?)and 0.56(?)) (P<0.05), but no statistical difference of the three sets of indexes was found between industrial group and medical group (P>0.05). (2) Analysis of the radiation workers with different genders: lymphocyte aberration sample positive rate, MF and CAF had no significant difference between male industrial group and female industrial group(7.58%vs6.94%, 1.85‰vs 1.83‰, 4.27(?)vs4.17(?)) (P>0.05), either was in medical group (7.83%vs9.09%, 1.58(?)vs1.83‰, 6.45(?)vs3.74(?)) (P>0.05). (3) Analysis of the radiation workers with different working ages: In industrial groups, lymphocyte aberration sample positive rates in 0 ?9 working age group, 10 ?19 working age group,20 ?29 working age group and above 30 working age group were 1.06%, 9.16%, 10.48%and 4.55% respectively. We found that lymphocyte aberration sample positive rates in 10?19 working age group and 20?29 working age group were significantly higher than that in 0?9 working age group (P<0.05). MFs in the four working age groups were 0.74‰, 1.94‰, 1.91‰ and 2.02‰ respectively. It was found that MFs in above 10 working age groups were significantly higher than that in 0 ?9 working age group(P<0.05). CAFs in the four working age groups were 0.53(?), 4.38(?), 6.67(?) and 5.68(?) respectively. We also found CAFs in above 10 working age groups were significantly higher than that in 0?9 working age group (P<0.05). In medical groups:lymphocyte aberration sample positive rates, MF and CAF in 0 ?9 working age group were 1.23%, 0.58‰, and 0.62‰, which were significantly lower than those in other three working age groups (10?19: 8.98%, 1.96(?),and 5.39‰,20?29: 12.22%, 1.99‰and 6.11(?), above 30: 10.61%, 2.04‰ and 9.09(?)) (P<0.05).Conclusions 1. Automatic scanning system of chromosome karyotype method was faster than Artificial method, it had lower missing rate and suited for large scale sample detections. Artificial method had higher missing rate and suited for little scale sample detections. Artificial method and Automatic scanning system of chromosome karyotype method had no obvious difference in peripheral blood lymphocytes aberration sample positive rates, the two methods shoule be applicated complementarily in detections. 2.The lymphocyte aberration sample positive rate, MF and CAF in radiation workers'peripheral blood were higher than those in non-radiation workers'.The contact of low-dose and long-term ionizing radiation can result in the peripheral blood lymphocytes'genetic changes in radiation workers, and the degree of changes increases with the working age. The professional protection of radiation workers need to be further strengthened.
