ABSTRCT Angiography and Expression of Vascular Endothelial Growth Factor and its receptors in the nude mice liver after planted human hepatocellular carcinoma cells In our country, the morbidity of human hepatocellular carcinoma is high, and, its mortality is very high, too. VEGF can be expressed in many human tumors, such as: astroblastoma, nasopharyngeal carcinoma, mammary cancer, carcinoma of kidney and so on. Lots of articles reporteded that VEGF could be overexpressed in hepatocarcinoma, and, that VEGFR resided in new tumor vascular endothelial cells mainly. But, the location of VEGF and its receptors and the mechanism of theirs are unclear. In our study, we watched the changes of hepatic vascular, especially, the changes of artery by angiography; we observed the pathologic changes of tumor location by HE staining; we investigated the expression of VEGF and its receptors by immunohistochemistry staining. We want to research the relationship between the genesis and advance of hepatocarcinoma and VEGF and its receptors. Materials and Methods twenty-five nude mice, 8 weeks old, 18-22g weighted. They were devided five groups, every group had five nude mice. The groups were control group, postoperation one-week group, postoperation two-week group and postoperation three-week group as well as postoperation four-week group. 10% pentobarbitone was ejected in nude mice abdomen. After five minutes, exposed the right lobe of theirs liver, rejected hepatic carcinoma cells 0.08-0.lml in them, the average number of cells were about 5 X 106. We make angiography, lIE staining and immunohistochemistry after postoperation one week, two weeks, three weeks and four weeks respectively. We ejected 76% meglucamine diatrizoate 0.Sml in one second from tail vein, at the same time, took photographies at the rate of four pieces per second. After angiography, we cut the chest wall to expose the heart, then, use a 6# pin to insert in aortic through left ventriculus. 3Oml physiological saline dropped in it quickly, followed, 4% paraformaldehyde was filled, then, the liver was cut off and was left in 40% paraformaldehyde to fix. Fixed liver was put in 2% glycogen to infiltrate about 24 hours, then, made frozen section. The thickness of tablet was about 2Oum. Followed, we make HE staining and immunohistochemistry. Result Tumor occurrence rate was 100% in postoperation groups. Tumor location was difficult to find by naked eyes in one-week group, but, tumor massae could be seen under light microscope. In two-week group, three livers could be seen tumor locations by naked eyes, and, the diametral was about 0.5-3.0mm, the number of locations was about 2-7. In three -week group, locations were more and larger than those of two -week group. On the other hand, normal liver structure was destroyed by light microscope. The locations of four-week group were the most and the largest, furthermore, we could find necrosis location by light microscope. Angiography: In three and four groups, new and irregular vascular was obvious. VEGF expression: Control group's expression was negative. The expression of one-week group was weak. The expression was high step by step from two-week to four-week group. VEGFR-1I fit-i expression: Ecs could express in control group.
