Effect of DHZCW on renal interstitial fibrosis and the association withexpressing of TGF- 13AbstractBackground and Objective: Renal interstitial fibrosis (RIF)is thecommon pathway to a series of chronic progressive renal disease. RIF is not only the pathological hallmark of chronic renal failure, but also one of the most important reason leading to end-stage renal disease. In kidney disease, it is the appearance of RIF, not that of glomirular injury meaning that this kidney is slowly going to progress. In end-stage, RIF can't deteriorated, and there aren't special treatment to RIF. But if RIF can be diagnosed early and be treated properly, RW may lightened and renal function will proved at same time. In this experiment, we observe the effect of Dahuangzhechongwan(DHZCW) in treating RIF rat model, meanwhile the expression of TGF--β1-the most important cytokine associating with organ fibrosis is also be tested. At last, we discussed the mechanism of DHZCW in treating RW, and we hope it may be a foundation for clinical using.Materials and Methods: 50 male SD rat were feed by 0.5% Adenine forage. After 8 weeks, the rats were identified to be RIF model when their blood urea(BUN), and serum creatinine (Scr) level reached to end-stage-5-of renal insufficiency. Then all the RIF model rats were randomly divided into Chinese traditional medicine group(CMG n= 12), west medicine group(WMG n=12) and control group(CG n=10). And all of them were respectively treated with DIIZCW, losrtan and water. 6 weeks later, SCr, BUN were tested, kidney biopsy was also taken and stained with PAS and Masson. The expression of TGF- f3 and TGF- f3 1mRNA were analyzed by immunohistochemistry and situ hybridization.Resluts:Compared with that in control group, in CHG and LG, theSCr and BUN had a sharp decrease, After treating, testing the value of serumScr, BUN and RilE area, in CMG they are 122.7 ?.53 i mol/L. 12.45?1.63mmol/L 0.4?.2; in WMG they are 134.5眑.59pmolIL 18.32?1.47mmol/L 0.6?.2; in CG they are 184.3?.63 t mol/L.. 23.41?1.52mmoI/L. 1.7?0.3 . The degree of renal interstitial fibrosis also lightened,meanwhile, the expression of TGF- 13 and TGF- 13 1mRNA decreased too.And those in CWG are more obvious.conclusion: DHZCW could prevent RIF by decreasing the fibrosis area, proving renal function. The main mechanism may be the reducing of the expressing of TGF- 13 1in renal.
