Objective An investigation on Platycodon grandifloroum(Jacq.)A.DC.cultivation history in Taihe Country, discuss its existing problems and develop strategies. To provide a standardized cultivation and field management of P.grandiflorum(Jacq.)A.DC., 16 kinds of inorganic elements and content distribution features of P.grandifloroum(Jacq.)A.DC.in Taihe were tested. To evaluate the quality of P.grandifloroum(Jacq.)A.DC.in Taihe whether stable, establishment of 12 sampels of HPLC fingerprint. To explore the differences about the content of platycodin D in different areas and different picked months.Methods A field survey of P.grandifloroum(Jacq.)A.DC.planting history in Taihe country, and access to literature. Microwave digestion for sample pretreatment, determined the content of elements in different parts and soil in different growth period by using ICP-MS, and analysis experiment results. Using HPLC-ELSD method established HPLC fingerprint of18 batches samples in different origin and 8 batches samples in different picked months,acetonitrile-0.1% formic acid as the mobile phase gradient, flow rate of 1.0m L ˙min-1,column temperature was 30℃, ELSD parameters for the nebulizer temperature was 70℃,the evaporator temperature was 90℃, N2 flow rate of 1.60 m L ˙ min-1. Platycodin D as control, determine 18 batches in different origin and 8 batches in different picked months of P.grandifloroum(Jacq.) A.DC..Results The elements from different regions have the same distributed, each element has a similar spectrum peak shape, and the content of elements descending order:K> Ca> Mg>Al> Fe> Mn(Ba)> Zn> Cr> Se> Mo, Mn and Ba elements in which no difference; there was more disparate of the same elements of the highest and the lowest content;Mg,Al, K, Ca, Fe, Se, Ba are typical characteristic elements of P.grandifloroum(Jacq.) A.DC.in Taihe. A total of 8 groups of elements was positively correlated; the content of elements show a certain regularity of P.grandifloroum(Jacq.) A.DC. root, stem and leaf in different harvest time; the enrichment ability of 16 kinds of elements in different picked months are different, enrichment values between 0.004 to 0.706, and the enrichment of Mg, K, Cu, Zn, Mo,Hg, Pb was strong. An HPLC fingerprint of P.grandifloroum(Jacq.)A.DC.contained 15 common peaks, and confirmed the platycodin D peak, of which 3batches of samples of similarity is less than 0.900; the content of chemical components changed significantly in 6, 8, 10, 11 month. The content of platycodin D was not stable in Taihe area; the content of platycodin D in different growth periods showed steady growth in the 4-8 month, reaching highest level in 9 month.Conclusion There is a long history of cultivation of P.grandifloroum(Jacq.)A.DC.before Qing dynasty in Taihe county. To provide references for agricultural standardized production of fertilizer, principal component, correlation and enrichment analysis of the content of inorganic elements P.grandifloroum(Jacq.)A.DC.in Taihe. This test establish the HPLC fingerprint and HPLC-ELSD method has exclusive, simple operation, good repeatability,and it provides a theoretical reference for the quality control of P.grandifloroum(Jacq.)A.DC.in Taihe.
